大豆卵磷脂对湖羊精液4℃保存效果的影响

摘要 为探究大豆卵磷脂(SL)对湖羊精液4℃保存效果的影响,在湖羊精液基础稀释液中添加不同浓度(0、0.1%、0.2%、0.3%和0.4%)的SL进行4℃低温保存试验,通过在不同保存时间(0~6 d)对精子活率、活力、直线速率、曲线速率、路径速率、质膜完整率和顶体完整率进行测定,确定湖羊精液低温保存稀释液中SL的最佳添加浓度。结果表明:保存2、5、6 d,0.1%SL组的精子活率、活力均显著高于其他组(P<0.05);保存3 d,0.1%SL组的精子直线速率显著高于其他组(P<0.05);保存4 d,SL各组的精子曲线速率和路径速率显著高于对照组(P<0.05);保存1、5 d,0.1%SL组的精子质膜完整率显著高于其他组(P<0.05);保存2、6 d,0.1%SL组的精子顶体完整率显著高于其他组(P<0.05)。结论:在低温保存条件下,添加0.1%SL可以有效地延长湖羊精子的存活时间,提高精液质量。 In order to investigate the effect of soy lecithin(SL)on the preservation of Hu sheep semen at 4℃,different concentrations(0,0.1%,0.2%,0.3%and 0.4%)of SL were added to the base dilution of Hu sheep semen for 4℃cryopreservation test.The optimum concentration of SL in the cryopreservation diluent of Hu sheep semen was determined by measuring sperm motility,viability,linear rate,curve rate,path rate,plasma membrane intactness and acrosome intactness at different points of the preservation time(0-6 days).The results showed that for 2,5 and 6 d of preservation,the sperm motility and vitality of the 0.1%SL group were significantly higher than those of the other groups(P<0.05);for 3 d of preservation,the linear rate of the 0.1%SL group was significantly higher than those of the other groups(P<0.05);for 4 d of preservation,the curve rate and path rate of the SL groups were significantly higher than those of the control group(P<0.05);for 1 and 5 d of preservation,the sperm plasma membrane integrity rate was significantly higher in the 0.1%SL group than in the other groups(P<0.05);and for 2 and 6 d of preservation,the sperm acrosome integrity rate was significantly higher in the 0.1%SL group than in the other four groups(P<0.05).In conclusion,under cryopreservation conditions,the addition of 0.1%SL effectively prolonged the survival time of spermatozoa and improved the quality of semen in Hu sheep.
