摘要
菌株Y5-11是从哈尔滨周边地区地下水中分离的一株假单胞菌,具有较高的自养脱氮能力。菌株Y5-11的16S rRNA基因序列与模式菌Pseudomonas koreensis Ps 9-14T高度同源(99.72%)。获得了菌株Y5-11的全基因组序列,采用比较基因组学分析了菌株Y5-11的碳源利用和脂肪酸含量特征,对菌株的生理生化特征进行了系统研究,发现菌株Y5-11与模式菌Pseudomonas koreensis Ps 9-14T在碳源利用上较为接近,但两株菌株在生长温度和硝酸盐还原方面存在差异。菌株Y5-11有17个特有基因,其在脂肪酸种类及含量上与其他菌株相比,存在明显的特异性,主要脂肪酸为C16:0和C17:0 cyclo,含量分别为19.37%和21.09%。通过以上研究,确定了菌株Y5-11的分类地位和生物学特征,为菌株Y5-11的进一步应用提供了生物学参考依据。
Pseudomonas sp.Y5-11 was isolated from groundwater in Harbin region and had the ability of autotrophic removal of nitrate.The 16S rRNA gene sequence of strain Y5-11 was found to be highly homologous to Pseudomonas koreensis Ps 9-14 T(99.72%).The complete genome sequences of the strain Y5-11 were obtained.The characteristics of carbon source utilization and fatty acid content of strain Y5-11 were analyzed by comparative genomics,and the physiological and biochemical characteristics of strain Y5-11 were systematically studied.It was found that strain Y5-11 was close to Pseudomonas koreensis Ps 9-14 T in carbon source utilization.However,there were differences in growth temperature and nitrate reduction between the two strains.The strain Y5-11 had 17 specific genes,which had obvious specificity in the type and content of fatty acids compared with other strains.The main fatty acids of strain Y5-11 were C16:0 and C17:0 cyclo with content of 19.37%and 21.09%,respectively.Through the above studies,the classification status and biological characteristics of strain Y5-11 were determined,which provided biological reference for further application of strain Y5-11.
