基于一测多评法的麸炒金樱子成分含量测定

Determination of components inbran rosa laevigata based on one-test-multiple-evaluation method

摘要 目的:建立一测多评法同时测定麸炒金樱子中没食子酸、芦丁、鞣花酸、山柰素、槲皮素、芹菜素、木犀草素7个化学成分的含量。方法:采用HPLC法测定,色谱柱:YMC Pack ODS-A C18(250 mm×4.6 mm,5μm);流动相:以甲醇为流动相A,0.1%磷酸溶液为流动相B,梯度洗脱(0~15 min,5%A;15~30 min,5%A~40%A;30~55 min,40%A~55%A;55~60 min,55%A~5%A;60~65 min,5%A),流速为1.0 mL·min^(-1),检测波长为272 nm,柱温为30℃。以鞣花酸为内参物,计算其他各成分的相对校正因子,对各成分进行定量分析,并与外标法测定结果进行比较。结果:没食子酸、芦丁、鞣花酸、槲皮素、木犀草素、山柰素、芹菜素分别在1.84~18.39、10.20~101.95、11.64~116.38、1.04~10.42、0.98~9.84、1.24~12.43、0.57~5.67μg·mL^(-1)范围内线性关系良好;平均加样回收率分别为94.60%、93.38%、91.42%、91.25%、92.70%、93.68%、92.66%;RSD分别为1.1%、0.91%、1.2%、1.6%、1.3%、1.2%和1.8%。结论:一测多评法同时测定麸炒金樱子中7个成分含量可操作性强且结果准确,可用于麸炒金樱子的质量控制。 OBJECTIVE To establish a one test multiple evaluation method for the simultaneous determination of seven chemical components:gallic acid,rutin,ellagic acid,kaempferol,quercetin,apigenin,and luteolin in roasted cherry blossoms with bran.METHODS Determined by HPLC,using methanol as the mobile phase A and 0.1%phosphoric acid solution as the mobile phase B,Gradient elution(0-15 min,5%A;15-30 min,5%A-40%A;30~55 min,40%A-55%A,55~60 min,55%A-5%A,60-65 min,5%A);Chromatographic column:YMC Pack ODS-A C18(250 mm×4.6 mm,5μm);The flow rate was 1.0mL·min^(-1);The detection wavelention was 272 nm;the column temperature was 30℃.Taking ellagic acid as internal reference material,the relative correction factors of other components were calculated,and the Quantitative analysis results were compared with those of external standard method.RESULTS The linear relationships of gallic acid,rutin,ellagic acid,quercetin,luteolin,Kaempferol and apigenin were in the range of 1.84-18.39、10.20-101.95、11.64-116.38、1.04-10.42、0.98-9.84、1.24-12.43、0.57-5.67μg·mL^(-1) respectively.The mean recoveries were 94.60%,93.38%,91.42%,91.25%,92.70%,93.68%and 92.66%;and the RSD were 1.1%,0.91%,1.2%,1.6%,1.3%,1.2%and 1.8%respectively.CONCLUSION The simultaneous determination of seven components in the seeds by one-test and multiple-evaluation method is Operability and accurate,and can be used for quality control of the seeds.
