CXCL1 promotes the proliferation and metastasis of cervical cancer cells by regulating JAK/STAT signaling pathway

目的探究CXCL1在宫颈癌中的表达及对宫颈癌细胞增殖、迁移和侵袭的影响及其作用机制。方法使用GEPIA在线数据库分析CXCL1在宫颈癌组织和正常组织中的表达水平,分析其与宫颈癌患者总生存期的关系。qRT-PCR检测CXCL1 mRNA在宫颈癌细胞(C-33A和Hela细胞系)和正常宫颈上皮细胞系(HcerEpic细胞系)中的表达。将CXCL1过表达质粒转染进宫颈癌细胞系中,CCK-8法检测CXCL2对宫颈癌细胞增殖的影响,TUNEL实验检测CXCL1对宫颈癌细胞凋亡的影响,Transwell实验检测CXCL1对宫颈癌细胞迁移和侵袭的影响,Western blot实验检测CXCL2对宫颈癌细胞系中p-JAK2和p-STAT3蛋白表达的影响。结果CXCL1在宫颈癌组织中的表达水平显著高于正常组织,且高表达CXCL1与宫颈癌患者总生存期较低有关。高表达CXCL1可显著促进宫颈癌细胞增殖,迁移和侵袭,并抑制细胞凋亡;促进p-JAK2和p-STAT3蛋白的表达。结论CXCL1通过调控JAK2/STAT3信号通路促进宫颈癌细胞增殖和转移,并抑制细胞凋亡。

Objective To explore the expression of CXCL1 in cervical cancer and its effect on the proliferation,migration and invasion of cervical cancer cells.Methods The expression of CXCL1 in cervical cancer tissues and normal tissues was analyzed by using GEPIA online database,and the relationship between CXCLI and overall survival of cervical cancer patients was analyzed.qRT-PCR was used to detect the expression of CXCL1 mRNA in cervical cancer cells(C-33A and Hela cell lines)and normal cervical epithelial cells(HcerEpic cellines).CXCL1 over-expression plasmid was transfected into cervical cancer cell lines.The effect of CXCL1 on the proliferation of cervical cancer cells was detected by CCK-8 method.TUNEL assay was used to detect the effect of CXCL1 on the apoptosis of cervical cancer cells.The effect of CXCL1 on the migration and invasion of cervical cancer cells was examined by Transwell assay.The expression of p-JAK2 and p-STAT3 in cervical cancer cell line was detected by Western blot.Results The expression level of CXCL1 in cervical cancer tissues was significantly higher than that in normal tissues,and the high expression of CXCL1 was associated with lower overall survival of cervical cancer patients.High expression of CXCL1 could significantly promote the proliferation,migration and invasion of cervical cancer cells,and inhibit apoptosis.High expression of CXCL1 could promote the expressions of p-JAK2 and p-STAT3.Conclusion CXCL1 can promote the proliferation and metastasis of cervical cancer cells and inhibit apoptosis by regulating JAK/STAT signaling pathway.
