甘草苷对口腔鳞状细胞癌细胞磷脂酰肌醇-3-羟激酶/蛋白激酶B/雷帕霉素靶蛋白信号通路的影响

摘要 目的 探讨甘草苷对口腔鳞状细胞癌(OSCC)细胞磷脂酰肌醇-3-羟激酶(PI3K)/蛋白激酶B(AKT)/雷帕霉素靶蛋白(MTOR)信号通路的影响。方法 提取114例OSCC患者的肿瘤细胞进行体外培养,调整细胞状态至对数生长期备用。采用CCK8法检测不同浓度(0、10、20、30、40μmol/L)甘草苷提取液对对数生长期的OSCC细胞存活率的影响;采用蛋白质印迹法(Western blot)检测OSCC细胞中PI3K、AKT、MTOR蛋白的相对表达量;采用逆转录聚合酶链反应(RT-PCR)检测PI3K、AKT、MTOR mRNA的相对表达量。结果 随着甘草苷提取液浓度的逐渐增加,OSCC细胞存活率逐渐下降,PI3K、AKT、MTOR蛋白及mRNA相对表达量均逐渐下降(P﹤0.05)。结论 甘草苷可能通过抑制OSCC细胞PI3K、AKT、MTOR蛋白及mRNA的表达抑制OSCC细胞生长,进而对OSCC产生抑制效果。 Objective To investigate the effect of liquiritin on the phosphatidylinositol 3-hydroxy kinase(PI3K)/protein kinase B(AKT)/mechanistic target of rapamycin kinase(MTOR) signaling pathway in oral squamous cell carcinoma(OSCC) cells.Method A total of 114 tumor cells from OSCC patients were extracted and cultured in vitro,and the cell state was adjusted to the logarithmic growth phase for later use.The effect of different concentrations(0,10,20,30,40 μmol/L) of liquiritin extract on the survival rate of OSCC cells in the logarithmic growth phase was tested by the CCK8 method.The relative expression levels of PI3K,AKT,and MTOR proteins in OSCC cells were detected by the Western blot method.The relative expression levels of PI3K,AKT,and MTOR mRNA were detected by reverse transcription-polymerase chain reaction(RT-PCR).Result As the concentration of liquiritin extract gradually increased,the survival of OSCC cells gradually decreased,and the relative expressions of PI3K,AKT,MTOR proteins and mRNA gradually decreased(P<0.05).Conclusion Liquiritin could suppress the growth of OSCC cells by inhibiting the expression of PI3K,AKT,MTOR proteins and mRNA in OSCC cells,thereby producing anti-tumor effects on OSCC.
