益骨汤对骨质疏松大鼠骨组织蛋白表达的影响

摘要 目的益骨汤是临床上用于治疗骨质疏松症的中药复方。然而,其治疗绝经后骨质疏松症的作用机制尚不清楚,需要进一步深入研究。方法使用TMT定量蛋白质组学对从模型组和益骨汤组大鼠骨组织中提取的蛋白质进行蛋白质组学分析,确定益骨汤干预后的差异表达蛋白以鉴定感兴趣的生物学相关蛋白,同时进行功能注释相关分析以探究其生物学功能及作用机制。结果GO功能富集分析得到GO条目156个,主要包括骨骼肌功能、酶活性和横纹肌作用;KEGG富集显示,矿物质吸收通路被显著富集。PPI蛋白质互作关系表明,相关蛋白可能是通过影响骨骼肌从而影响了骨质疏松的发生。结论益骨汤对绝经后骨质疏松治疗有效果。其中,益骨汤的作用机制可能是通过肠道影响矿物质吸收从而影响了相关蛋白的表达,进而调控绝经后骨质疏松的发生和发展。 Objective Yigu decoction is a compound Chinese medicine used to treat osteoporosis in clinic.However,its mechanism of action in the treatment of postmenopausal osteoporosis is not clear and needs further study.Methods Protein extracts from bone tissues of rats in the estradiol valerate group and the YGD group were subjected to proteomic analysis using TMT quantitative proteomics to identify differentially expressed proteins after YGD and estradiol valerate interventions to identify biologically relevant proteins of interest,and functional annotation correlation analysis was performed to explore their biological functions and mechanisms of action.Results GO functional enrichment analysis yielded 156 GO entries,mainly including skeletal muscle function,enzyme activity and transverse muscle action.KEGG enrichment showed that the mineral uptake pathway was significantly enriched.PPI protein interactions indicated that the relevant proteins might have influenced the development of osteoporosis by affecting skeletal muscle and thus osteoporosis.Conclusion Both YGD and estradiol valerate are effective in the treatment of postmenopausal osteoporosis.The mechanism of action of YGD may be to influence mineral absorption through the intestine,thus affecting the expression of the related proteins and regulating the development of postmenopausal osteoporosis.
