摘要
目的:基于网络药理学探讨柴胡桂枝干姜汤治疗腺样体肥大的作用机制。方法:通过中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP),BATMAN-TCM数据库结合口服利用度(oral bioavailability,OB)、类药性(drug-likeness,DL),药物半衰期(drug half-life,HL)检索柴胡桂枝干姜汤中柴胡、桂枝、干姜、黄芩、瓜蒌根、牡蛎、炙甘草的候选化合物及作用靶点信息,并通过Uniprot数据库将靶点校正为基因名;运用Genecards数据库获取腺样体肥大的相关靶点与基因,利用VENNY图获得柴胡桂枝干姜汤与腺样体肥大的交集靶点;STRING数据库来构建蛋白质相互作用PPI网络图;David数据库对交集靶点进行基因本体(gene ontology,GO)功能富集分析和KEGG通路富集分析;Cytoscape软件进行网络靶点特性分析。结果:筛选出柴胡桂枝干姜汤候选化合物65个,转化为有效靶点288个,得到腺样体肥大相关靶点781个,匹配得到共有靶点118个。GO富集主要涉及597个生物学过程、60个细胞组成和106个分子功能,KEGG通路分析主要涉及112条通路。结论:柴胡桂枝干姜汤治疗腺样体肥大是通过多靶点多通路发挥作用的。
Objective:To explore the mechanism of treating adenoid hypertrophy by Chaihu Guizhi Ganjiang Tang based on network pharmacology.Methods:TCMSP and BATMAN-TCM databases were used in combination with OB,DL and HL,to search for candidate compounds and targets of Chaihu(Bupleuri radix),Guizhi(Cinnamomi ramulus),dried ginger,Huangqin(Scutellariae radix),Tianhuafen(Trichosanthis radix),Muli(Ostreae concha)and Zhigancao(Glycyrrhizae radix et rhizoma praeparata cum melle)in the decoction,and the targets were corrected to the names of genes via the Uniprot database;Genecards was applied to obtain the targets and genes related to adenoid hypertrophy,Venny diagram was utilized to obtain the intersection targets of Chaihu Guizhi Ganjiang Tang and adenoid hypertrophy;STRING database to construct PPI network of protein interaction;David database to perform GO function enrichment analysis and KEGG enrichment analysis of the intersection targets;Cytoscape software to analyze the characteristics of network targets.Results:All 65 candidate compounds of Chaihu Guizhi Ganjiang Tang were screened out,and transformed into 288 active targets,obtaining 781 adenoid hypertrophy-related targets and 118 common targets matched.GO enrichment mainly involved 597 biological processes,60 cellular compositions and 106 molecular functions,and KEGG pathway analysis mainly dealt with 112 pathways.Conclusion:Treatment of adenoid hypertrophy by Chaihu Guizhi Ganjiang Tang could develop the effects by multi-target and multi-pathway.
