摘要
[目的]通过泊洛沙姆407(Poloxamer 407,P-407)诱导建立一种新型多器官脂肪变性模型。[方法]将50只雄性SD大鼠按随机数表法分成对照组(0.9%生理盐水)、Ⅰ组(0.25 g/kg P-407)、Ⅱ组(0.50 g/kg P-407)、Ⅲ组(1.00 g/kg P-407)、Ⅳ组(2.00 g/kg P-407),连续注射3个月后,检测血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)水平,并取大鼠心、肝、脾、肺、肾、胰等脏器组织进行固定、包埋、染色处理,观察各器官组织病理变化。[结果]与对照组相比,P-407组TC、TG、HDL、LDL水平明显升高,各组TC水平分别增高202%、470%、1260%、2068%(P<0.001);各组TG水平分别增高581%、1146%、2322%、4129%(P<0.001);各组HDL水平分别升高205%、406%、504%、716%(P<0.001);各组LDL水平分别升高797%、1513%、2135%、2870%(P<0.001);HE染色结果显示,P-407实验组肝、肾脏器发生脂肪变性,随着P-407浓度升高,脂肪变性程度逐渐增加,第Ⅳ组(2.00 g/kg P-407)脂肪变性程度最重。[结论]成功建立了一种新型的多器官脂肪变性模型,在2.00 g/kg P-407中,大鼠肝、肾脏器中出现的脂滴数目最多,各血清脂质水平升高最明显。
[Objective]To establish a novel multi-organ steatosis model induced by Poloxamer 407(P-407).[Method]Fifty male SD rats were divided into control group(0.9%saline),groupⅠ(0.25 g/kg P-407),groupⅡ(0.50 g/kg P-407),groupⅢ(1.00 g/kg P-407)and groupⅣ(2.00 g/kg P-407)according to the random number table method.The levels of serum triglyceride(TG),total cholesterol(TC),high-density lipoprotein(HDL)and low-density lipoprotein(LDL)were measured after three months.[Result]Compared with the control group,TC,TG,HDL,and LDL levels were significantly higher in the P-407 group,with TC levels increasing by 202%,470%,1260%,and 2068%in each group,respectively(P<0.001);TG levels increasing by 581%,1146%,2322%,and 4129%in each group,respectively(P<0.001);HDL levels increasing by 205%,406%,504%,716%(P<0.001);LDL levels increased by 797%,1513%,2135%,2870%(P<0.001)in each group,with statistically significant differences(P<0.001);HE staining results showed that steatosis occurred in liver and kidney organs in the P-407 experimental group,and the degree of steatosis gradually increased with the increase of P-407 concentration.The degree of steatosis gradually increased as the concentration of P-407 increased,and the degree of steatosis in groupⅣ(2.00 g/kg P-407)was the most severe.[Conclusion]A new model of multi-organ steatosis was established successfully.In the 2.00 g/kg P-407 group,the number of lipid droplets in the liver and kidney was the highest,and the level of serum lipid was the most obvious.
