枣PAO基因的鉴定及在果实发育和贮藏过程中的表达分析 被引量:1

摘要 多胺氧化酶(PAO)是一种黄素腺嘌呤二核苷酸依赖酶,催化多胺(PA)分解代谢,在植物的生长发育和胁迫反应过程中起着关键作用。本研究对枣PAO基因进行了全基因组筛选,共鉴定出3个ZjPAO基因(ZjPAO1、ZjPAO2、ZjPAO3),并对它们的保守结构域、基因结构、染色体位置、进化关系、亚细胞定位、启动子顺式作用元件等进行预测和分析。结果表明,ZjPAO1、ZjPAO2、ZjPAO3基因分别分布在4、5、8号染色体上,结构特征较保守,都包含Amino-oxidase结构域。通过构建系统发育树,将拟南芥、番茄、桃和枣的PAO蛋白分为4类,并发现ZjPAOs蛋白与桃PAOs的亲缘关系较近。ZjPAOs基因启动子含有激素响应、应激响应等多种顺式作用元件,预测在枣生长发育中发挥作用。此外,转录组数据分析表明,ZjPAO1和ZjPAO2基因在枣果实不同发育阶段均有表达;在外源水杨酸处理下,ZjPAO1基因被显著诱导表达。本研究结果有助于了解延缓鲜枣果实成熟和衰老的机制。 Polyamine oxidase(PAO)is a flavin adenine dinucleotide dependent enzyme that catalyzes the catabolism of polyamines(PA)and plays a crucial role in plant growth,development and stress response.The genome-wide screening of PAO genes in jujube was conducted,and three ZjPAO genes(ZjPAO1,ZjPAO2,ZjPAO3)were identified,then their conserved domains,gene structures,chromosomal positions,evolutionary relationships,subcellular localization and promoter cis acting elements were analyzed.The results showed that the three ZjPAO genes were distributed on Chr4,Chr5 and Chr8,respectively,with relatively conservative structural characteristics and the Amino-oxidase domain.By constructing the phylogenetic trees of the PAO genes of Arabidopsis,tomato,peach and jujube,we found that they could be divided into four catego-ries,and the ZjPAO geneswere closely related to the PAO genes in peach.The promoter of the ZjPAO genes contained various cis acting elements related with such as hormones and stress responses,which were predicted to play roles in jujube growth and development.In addition,the transcriptome data analysis showed that both ZjPAO1 and ZjPAO2 expressed at different stages of jujube fruit development.Under exogenous salicylic acid treatment,the ZjPAO1 gene was significantly induced to express.The results of this study could contribute to understanding the mechanisms of delaying ripening and aging of fresh jujube fruits.
