摘要
前动力蛋白2(Prokinetic 2,PROK2)是一种含多个半胱氨酸的多功能蛋白,参与调节机体生理功能。PROK2与其受体前动力蛋白受体1/2(Prokineticin receptor 1/2,PKR1/2)在中枢神经系统中广泛存在,病理损伤可增加受损处或血清中PROK2的表达水平,表明PROK2可能参与多种中枢神经系统疾病的发病过程。目前研究表明高表达PROK2对中枢神经系统疾病同时具有保护和损伤双重作用,具体作用机制尚未完全研究清楚。本文结合近年来国内外研究文献,从PROK2对多种中枢神经系统疾病的影响进行综述。文献复习表明,PROK2参与了中枢神经系统疾病的发生发展,包括缺血性脑卒中、蛛网膜下腔出血、创伤性脑损伤、帕金森病、阿尔茨海默病、亨廷顿病、多发性硬化症、癫痫和嗅觉异常等,而PKR的拮抗剂已被证实可减缓中枢神经系统疾病的进展。这些结果表明PROK2可能是治疗中枢神经系统疾病的关键靶点,为治疗神经系统疾病的研究方向提供参考。
Prokinetic 2(PROK2)is a multifunctional protein containing multiple cysteines,which is involved in the regulation of physiological functions in the body.PROK2 and its receptor 1/2(PKR1/2)are widely present in the central nervous system,and pathological injury can increase the expression level of PROK2 at the damaged site or in serum,suggesting that PROK2 may be involved in the pathogenesis of a variety of central nervous system diseases.Current studies have shown that high expression of PROK2 has both protective and damaging effects on central nervous system diseases,and its specific mechanism has not been fully studied.This article reviews the neuroprotection and neurodamage of PROK2 in various central nervous system diseases.The literature review results show that PROK2 is involved in the occurrence and development of central nervous system diseases,including ischemic stroke,subarachnoid hemorrhage,traumatic brain injury,Parkinson's disease,Alzheimer's disease,Huntington's disease,multiple sclerosis,epilepsy,and dysolfactory disorders.Antagonists of PKR have been shown to slow the progression of central nervous system diseases.These results indicate that PROK2 is a key target for the treatment of central nervous system diseases,and provide a reference for the research direction of the treatment of nervous system diseases.
