不同油脂体系对天然抗氧化剂透皮吸收研究

摘要 研究在不同不饱和脂肪酸含量油脂组成的微乳体系中天然抗氧化剂透皮吸收特性。采用Franz扩散仪,以离体猪皮作为透皮屏障,在含有不同不饱和脂肪酸含量油脂制成的微乳体系中加入天然的α-维生素E、α-维生素E醋酸酯作为样本,用高效液相色谱法测定其在24 h内不同时间段接收液透皮量和24 h后猪皮角质层中活性成分,通过数据分析,比较其透皮吸收性能。由实验结果可知,通过D相微乳化技术成功制备乳液体系在室温下稳定性良好,活性成分体外透皮速率要比纯油脂中高,更容易渗透入皮肤;山茶籽油不饱和脂肪酸含量≥90%,且不饱和脂肪酸中油酸含量占比最高,油酸可以影响脂质层结构流动,因此其山茶籽油组成的微乳体系中天然抗氧化剂α-维生素E、α-维生素E醋酸酯透皮效果优于其他体系,且容易滞留在皮肤表皮层。 The transdermal absorption of natural antioxidants in the reverse microemulsions containing different oils was studied.The samples were prepared by adding naturalα-vitamin E andα-vitamin E acetate into the reverse microemulsions prepared with different oils.Using Franz diffusion apparatus,taking isolated pig skin as a transdermal barrier,the permeation quantities in different receptor solutions in 24 h and the intradermal retention after 24 h were measured by HPLC.The transdermal absorption properties were compared.The results showed that,the emulsion system successfully prepared by D-phase microemulsification technology had good stability at room temperature,and the transdermal rate of active ingredients was higher than that of pure oil,which was easier to penetrate into the skin.The content of unsaturated fatty acids in camellia seed oil was≥90%,and the proportion of oleic acid was the highest among the unsaturated fatty acids.Oleic acid could affect the flow of lipid layer structure.Therefore,the transdermal effects of natural antioxidantsα-vitamin E andα-vitamin E acetate in the microemulsion system composed of camellia seed oil were better than other systems,and the two natural antioxidants therein were easy to stay in the epidermal layer of the skin.
