摘要
2型糖尿病是全球增长速度最快的代谢性疾病之一,据预测到2040年糖尿病患者将增加到6.42亿,而其中30%~40%会发展为糖尿病肾病。胰岛素样生长因子-1(IGF-1)是一种由70个氨基酸组成的多肽,与胰岛素具有同源性,具有类胰岛素作用,又在细胞增殖、分化、凋亡以及机体的代谢、胚胎发育、肿瘤发生和发展等过程中发挥着重要作用。近年来众多研究发现,IGF-1直接或间接参与了糖尿病的病理生理过程,并且通过胰岛素相关信号通路与糖尿病肾病的发生、发展存在紧密联系。环状甘氨酸—脯氨酸(cGP)是IGF-1的代谢产物,参与IGF-1的调节,cGP/IGF-1代表有生物活性的IGF-1。现综述IGF-1在2型糖尿病及2型糖尿病肾病中的可能作用,为其治疗和预防提供新思路。
Type 2 diabetes is one of the fastest growing metabolic diseases in the world.It is predicted that the number of diabetes patients will increase to 642 million by 2040,of which 30%~40%will develop into diabetes nephropathy.Insulin-like growth factor-1(IGF-1)is a peptide composed of 70 amino acids,which is homologous to insulin and has insulin-like effects.It also plays an important role in cell proliferation,differentiation,apoptosis,metabolism,embryonic development,tumor occurrence,and development.In recent years,many studies have found that IGF-1 directly or indirectly participates in the pathophysiological process of diabetes,and is closely related to the occurrence and development of diabetes nephropathy through insulin related signaling pathways.Cyclic glycine proline(cGP)is a metabolite of IGF-1 and participates in the regulation of IGF-1.cGP/IGF-1 represents the biologically active IGF-1.This article reviews the possible role of IGF-1 in type 2 diabetes and type 2 diabetes nephropathy,and provides new ideas for its treatment and prevention.
