全自动免疫磁珠法与免疫亲和柱法测定玉米粉中黄曲霉毒素B_(1)的对比研究

摘要 采用全自动免疫磁珠净化与免疫亲和柱净化的前处理方法,结合超高效液相色谱法对玉米粉中黄曲霉毒素B_(1)进行定量检测,并对两种前处理方法结果准确性、加标回收率、检测时间进行了对比。结果表明:对玉米粉中添加不同量的黄曲霉毒素B_(1)标准物质,使用免疫亲和柱净化法的加标回收率在80.96%~100.42%之间,平均回收率为88.04%,标准偏差为0.052,变异系数为5.90%;使用全自动免疫磁珠净化法的加标回收率在90.00%~108.33%之间,平均回收率为100.20%,标准偏差为0.046,变异系数为4.57%。采用配对t检验评价两种净化方式对检测结果一致性的影响,结果显示t值小于理论t_(0.05(32))的值,P>0.05,结果显示这两种净化方法的差异在可接受范围内。两种方法在净化玉米粉中黄曲霉毒素B_(1)中可以满足实验要求。全自动免疫磁珠净化搭配使用真菌毒素全自动净化仪前处理方法可同时处理多个样品,平均一个样品净化时间小于3 min,处理时间短、实验效率更高。 The pretreatment methods of automatic immunomagnetic bead purification and immunoaffinity column purification were combined with ultra-high performance liquid chromatography to quantitatively detect aflatoxin B1 in corn flour,and the accuracy,spiked recovery and detection time of the two pretreatment methods were compared.The results showed that the recovery rate of aflatoxin B1 standard substance in corn flour was 80.96%~100.42%,the average recovery rate was 88.04%,the standard deviation was 0.052,and the coefficient of variation was 5.90%.The recovery rate of the automatic immunomagnetic bead purification method was 90.00%~108.33%,the average recovery rate was 100.20%,the standard deviation was 0.046,and the coefficient of variation was 4.57%.Paired t test was used to evaluate the effect of the two purification methods on the consistency of the test results,the results showed that the t value was less than the theoretical t_(0.05(32)) value,P>0.05,this inducated that the difference between this two purification methods was within the acceptable range.The two methods can meet the experimental requirements in purifying aflatoxin B1 in corn flour.The automatic immunomagnetic bead purification combined with the pretreatment method of the mycotoxin automatic purification instrument could simultaneously process multiple samples.The average purification time of a sample was less than 3 min,and the processing time was shorter and the experimental efficiency was higher.
