摘要
目的对三七、灯盏花、天麻、水蛭、龙血竭抗缺血性脑卒中进行对比实验研究,为确定组方药材提供试验依据。方法通过断尾法和毛细管法分别测定小鼠尾出血时间(BT)、毛细管凝血时间(CT);采用ADP诱导小鼠急性肺血栓模型和角叉菜胶诱导小鼠慢性血栓模型,观察药物对血栓形成的影响;采用小鼠断头缺血模型,观察药物对小鼠呼吸次数及呼吸持续时间的影响。结果除天麻提取物外,其余所有给药组均能显著延长小鼠的出血时间,其效果以阳性药ASP最优。三七总皂苷的凝血效果最佳。灯盏细辛对抗ADP诱导的小鼠肺血栓形成的作用最显著。对角叉菜胶诱导小鼠的尾部血栓形成模型三七总皂苷低剂量组比高剂量组好。结论各样品均有显著抗凝、抗血栓及抗脑缺血缺氧作用,但侧重点有所差异。
Objective:To make a contrast experiment study on the antithrombotic effects of Panax notoginseng,Erigeron breviscapus,Gastrodia elata,Leech and Resina Draconis on ischemic stroke,and to provide experimental basis for determining the prescription component.Methods:The tail bleeding time(BT)and the capillary coagulation time(CT)were measured by tail breaking method and capillary method respectively.ADP induced mice acute pulmonary thrombosis model and carrageenan induced mice chronic thrombosis model were used to observe the effects of the medicine on thrombosis.The ischemic model of severed head mice was used to observe the effects of the medicine on the number of breaths and the duration of breaths in mice.Results:Except for Gastrodia extract,all the other administration groups could significantly prolong the bleeding time of mice,and the positive ASP was the best.The total saponins of Panax notoginseng had the best coagulation effect.The effect of Breviscapus on ADP-induced pulmonary thrombosis in mice was most significant.The tail thrombus formation model of mice induced by carrageenann was better in the low dose group than in the high dose group.Conclusion:All the samples have significant anticoagulation,antithrombotic and anticerebral ischemia and hypoxia effects,but the emphasis is different.
