老年克罗恩病血清lncRNA MALAT1和LUCAT1表达情况及其与肠道菌群数量的相关性

摘要 目的探讨老年克罗恩病(CD)患者血清lncRNA MALAT1和LUCAT1表达及其与肠道菌群数量的相关性。方法选取2021年1月~2022年1月浙江省丽水市第二人民医院收治的老年CD患者120例为CD组,另外选择同期正常健康体检者80例为对照组。应用qRT-PCR法检测血清中lncRNA MALAT1、LUCAT1表达水平;通过革兰染色对菌群鉴定并计算肠道菌群数量;采用Pearson相关性分析血清lncRNA MALAT1、LUCAT1表达水平与肠道菌群数量的相关性。结果(1)CD组血清中lncRNA MALAT1的表达水平显著低于对照组(P<0.01),LUCAT1的表达水平显著高于对照组(P<0.01);lncRNA MALAT1和LUCAT1CD在CD组三个不同活动期中差异有统计学意义(P<0.01),其中,重度活动期血清lncRNA MALAT1的表达水平显著低于中度活动期和缓解期,中度活动期表达水平显著低于缓解期(P<0.01);重度活动期血清LUCAT1的表达水平显著高于中度活动期和缓解期,中度活动期表达水平显著高于缓解期(P<0.01)。(2)相关性分析显示,缓解期、中度活动期和重度活动期CD患者血清lncRNA MALAT1的表达水平与普氏菌、双歧杆菌的数量呈正相关,与肠球菌、大肠杆菌的数量呈负相关(P<0.01);LUCAT1表达水平与普氏菌、双歧杆菌的数量均呈负相关,与肠球菌、大肠杆菌的数量呈正相关(P<0.01)。结论CD患者血清lncRNA MALAT1表达水平显著降低,而LUCAT1表达水平显著增高,两者与肠道菌群数量有相关性。 Objective To investigate the expression of serum lncRNA MALAT1 and LUCAT1 in elderly patients with Crohn’s disease(CD)and their correlation with the number of gut microbiota.Methods 120 elderly CD patients admitted to the Second People’s Hospital of Lishui City,Zhejiang Province from January 2021 to January 2022 were selected as the CD group,and 80 healthy individuals who underwent physical examinations during the same period were selected as the control group.Using qRT PCR method to detect the expression levels of lncRNA MALAT1 and LUCAT1 in serum;Identify the microbiota through Gram staining and calculate the number of gut microbiota;Pearson correlation analysis was used to investigate the correlation between the expression levels of serum lncRNA MALAT1 and LUCAT1 and the number of gut microbiota.Results(1)The expression level of lncRNA MALAT1 in the serum of the CD group was significantly lower than that of the control group(P<0.01),while the expression level of LUCAT1 was significantly higher than that of the control group(P<0.01);There was a statistically significant difference in the expression levels of lncRNA MALAT1 and LUCAT1CD among the three different activity stages in the CD group(P<0.01).Among them,the expression levels of lncRNA MALAT1 in the serum during the severe activity stage were significantly lower than those during the moderate activity stage and remission stage,and the expression levels in the moderate activity stage were significantly lower than those in the remission stage(P<0.01);The expression level of serum LUCAT1 in severe active phase was significantly higher than that in moderate active phase and remission phase,and the expression level in moderate active phase was significantly higher than that in remission phase(P<0.01).(2)Correlation analysis showed that the expression level of serum lncRNA MALAT1 in patients with CD in remission,moderate activity,and severe activity stages was positively correlated with the number of Prevotella and Bifidobacterium,and negatively correlated with the number of Enterococcus and Escherichia coli(P<0.01);The expression level of LUCAT1 was negatively correlated with the number of Prevotella and Bifidobacterium,and positively correlated with the number of Enterococcus and Escherichia coli(P<0.01).Conclusion The expression level of serum lncRNA MALAT1 in CD patients is significantly reduced,while the expression level of LUCAT1 is significantly increased,which is correlated with the number of gut microbiota.
