改性C18色谱柱对三七总皂苷含量测定的影响

摘要 改性C18色谱柱是在C18结构中添加苯基后,大幅改变C18色谱柱对特定化合物的分离特性,文章通过三七总皂苷中人参皂苷Re的改变进行分析说明,将新构建的改性C18色谱柱与标准C18运行方法进行了多样品含量对比,并对重现性、标准曲线稳定性、加标回收率进行了研究。结果显示:该改性色谱柱大幅提升了人参皂苷Re分离度,并能缩短液相分析总时长;使用150 mm长度色谱柱就能够获得250 mm长度色谱柱的理论塔板数,所测含量结果偏差<2%。 The modified C18 chromatography column significantly changes the separation characteristics of specific compounds by adding benzene to the C18 structure.This article analyzes and explains the changes in ginsenoside Re in the total saponins of Panax notoginseng,compares the newly constructed modified C18 chromatography column with the standard C18 operating method for multiple sample content,and studies the reproducibility,stability of the standard curve,and recovery rate.The results showed that the modified chromatographic column significantly improved the separation of ginsenoside Re and shortened the total liquid phase analysis time.By using a 150mm length chromatographic column,the theoretical number of plates for a 250mm length chromatographic column can be obtained,and the deviation of the measured content results is less than 2%.
