摘要
以rhIL 11作为免疫原常规免疫BALB/c小鼠 ,采用细胞融合、ELISA检测和杂交瘤细胞克隆筛选等方法获得了 2株鼠抗人IL 11的单克隆抗体 (5A4、 1G12 ) ,分别属于IgG1亚类和IgG2a亚类。MTT法和细胞增殖试验分析表明 ,5A4、 1G12均不同程度地抑制IL 11对其依赖株细胞B9 11的增殖效应。表明成功地研制成了 2株鼠抗人IL
RhIL-11 was used as immunogen to immunize BALB/c mouse Two hybridoma cell lines,which secrete specific McAb against human IL-11,were obtained by using cell fusion,limited coloning assay,and ELISA screening The specificity of McAb was verified by Western blotting and the subsets of IgG,which the McAb belong to,were indicated by quickly test-paper analysis The biological function of the McAb in the proliferation of B9-11 cells was measured by MTT assay in vitro Two anti-human IL-11 McAb,5A2 and 1G12,which is belonging to IgG1 and IgG2 subset respectively,were obtained Both of them can inhibit the proliferation of an IL-11-dependent cell line,B9-11 cell line respectively,when IL-11 was present in culture system Thus two neutralizing anti-human IL-11 specific McAb were successfully obtained -
