基于网络药理学探究栀子治疗高脂血症的作用

摘要 目的:借助网络药理学探究栀子治疗高脂血症的作用机制。方法:借助TCMSP、Uniprot、SwissTargetPrediction数据库获取栀子的主要成分和潜在作用靶点;通过Genecards数据库检索出高脂血症基因;在Venny 2.0.1平台获取交集基因,然后将其导入String数据库进行分析,利用Cytoscape 3.9.1软件绘制出“药物-成分-共同基因”网络;使用String数据库构建出PPI网络,然后利用David数据库进行GO功能富集分析和KEGG通路富集分析。结果:筛选出栀子治疗高脂血症的核心成分为槲皮素、齐墩果酸、山柰酚、5-羟基-7-甲氧基-2-(3,4,5-三甲氧基苯基)色酮等,核心靶点包括RXRA、AKT1、MAPK14等,治疗高脂血症涉及的主要通路有癌症通道、脂质和动脉粥样硬化、糖尿病并发症中的AGE-RAGE信号通路、人类巨细胞病毒感染、流体剪切应力与动脉粥样硬化等。结论:栀子通过多种成分、多个靶点以及多条通路对高脂血症进行治疗。 Objective:The mechanism of gardenia in treating hyperlipidemia was investigated by means of network pharmacology.Method:The main components and potential targets of gardenia gardenia were obtained by TCMSP,Uniprot and SwissTargetPrediction database.The hyperlipidemia gene was retrieved from Genecards database.The intersection gene was obtained on Venny 2.0.1 platform,and imported into String database for analysis.The drug-components-common gene network was mapped using Cytoscape 3.9.1 software.The PPI network was constructed using String database,and then the GO function enrichment analysis and KEGG path enrichment analysis were performed using David database.Result:The core components of gardenia in the treatment of hyperlipidemia were quercetin,oleanolic acid,kaempferol,5-hydroxy-7-methoxy-2-(3,4,5-trimethoxyphenyl)chromone,etc.The core targets included RXRA,AKT1,MAPK14,etc.The main pathways involved in the treatment of hyperlipidemia include cancer pathways,lipids and atherosclerosis related pathways,AGE-RAGE signaling pathway in diabetes complications,human cytomegalovirus infection related pathways,fluid shear stress and atherosclerosis related pathways.Conclusion:Gardenia can treat hyperlipidemia through multiple components,multiple targets and multiple pathways.
