摘要
目的 探索肠道菌群在麻杏石甘汤治疗肺炎中的作用机制。方法 采用脂多糖雾化吸入法制备大鼠肺炎损伤模型,通过高通量测序和代谢组学检测动物肠内容物,分析麻杏石甘汤干预后动物肠道菌群和代谢产物的变化;以组学和高通量映射的KEGG通路为核心,联合分析代谢组和高通量测序结果。结果 经麻杏石甘汤治疗后,动物肺组织肺损伤情况显著好转。在肠道内容物中,优势菌种有Staphylococcaceae、Gemellaceae、Lactobacillaceae等,含量显著上调的代谢产物有芥酸、琥珀酸半醛、二十碳二烯酸等。其中Veilonella、Aerococcus等菌与磷酸氨基甲酸、琥珀酸半醛、维生素D2、棕榈酸、马来酸等相关性较为显著。结论 麻杏石甘汤可以缓解肺组织炎症损伤,其作用机制与通过肠道菌群调节棕榈油酸、马来酸、磷酸、1-甲基组氨酸、3-脱氢莽草酸等代谢产物有关。
Objective To explore the mechanism of action of intestinal flora in the treatment of pneumonia with Maxing Shigan Decoction.Methods A rat model of pneumonia injury was prepared by LPS aerosol inhalation method,and the intestinal content was tested by high-throughput sequencing and metabolomics.The changes in intestinal flora and metabolic products were analyzed after Maxing Shigan Decoction intervention;the KEGG pathway was used as the core for combined analysis of metabolomics and high-throughput sequencing results.Results After treatment with Maxing Shigan Decoction,the animal lung tissue injury was significantly improved.In the intestinal content,dominant bacteria included Staphylococcaceae,Gemellaceae,Lactobacillaceae,etc.,and the content of metabolites such as Erucic acid,Succinic acid semial,Eicosadienoic acid was significantly up-regulated.Among them,Veilonella,Aerococcus and other bacteria have a significant correlation with phosphocarbamate,succinic acid hemialdehyde,vitamin D2,palmitic acid,maleic acid,etc.Conclusion Maxing Shigan Decoction can alleviate lung tissue inflammation and injury.Its mechanism of action is related to regulating palm oil acid,maleic acid,phosphoric acid,1-methylhistidine,3-dehydrogenated shikimate acid and other metabolites through the regulation of gut microbiota.
