齐墩果酸对晚期糖基化终产物加重链脲佐菌素诱导糖尿病肾病小鼠保护作用

Protection of Oleanolic Acid on STZ⁃induced Diabetic Nephropathy Aggravated by AGEs in Mice

目的探讨山茱萸中效应成分齐墩果酸对晚期糖基化终产物(AGEs)加重链脲佐菌素(STZ)诱导糖尿病肾病小鼠保护作用。方法小鼠随机分为空白对照组、AGEs组、模型组、齐墩果酸低剂量组、齐墩果酸高剂量组,每组8只。将STZ诱导成功的DN小鼠饲喂高AGEs饲料,并分别灌胃给予齐墩果酸低剂量(0.02 g·kg^(-1))和齐墩果酸高剂量(0.10 g·kg^(-1))。8周治疗后,血糖试纸检测血糖值(Glu),试剂盒检测血清胰岛素(INS)、血清晚期糖基化终产物(AGEs)、血清肌酐(Scr)、血尿素氮(BUN)、尿肌酐(Cr)、尿白蛋白(UA)的水平。结果齐墩果酸可显著降低DN小鼠血糖,升高血清胰岛素、尿肌酐水平,降低血清AGEs、血清尿素氮和肌酐、尿蛋白水平。结论齐墩果酸对DN小鼠的血糖和肾脏具有一定保护作用,且高剂量作用较优。

OBJECTIVE To observe the protective effect of oleanolic acid(an active component in Cornus officinalis)on the diabetic nephropathy(DN)mice induced by streptozocin(STZ)and aggravated by advanced glycation end products(AGEs).METHODS Mice were randomly divided into blank control group,AGEs group,model group,oleanolic acid lowdose group and oleanolic acid highdose group,with 8 mice in each group.DN mice successfully induced by STZ were fed diets of high AGEs,and given low dose of oleanolic acid(0.02 g·kg^(-1))and high dose of oleanolic acid(0.10 g·kg^(-1))by intragastric administration.After 8 weeks of treatment,serum insulin(INS),serum advanced glycation end products(AGEs),serum creatinine(Scr),blood urea nitrogen(BUN),urinary creatinine(Cr),urinary albumin(UA)levels were measured by the kit.RESULTS Oleanolic acid could significantly reduce blood glucose,increase serum insulin and urinary creatinine levels,and decrease serum AGEs,serum urea nitrogen,creatinine and urinary protein levels in DN mice.CONCLUSION Oleanolic acid has the protactive effects on blood sugar and kidney of DN mice,and the high dose was much better.
