姜黄素联合二甲双胍对卵巢癌细胞SKOV-3凋亡的影响

Effects of Metformin Combined with Curcumin on Apoptosis of Ovarian Cancer SKOV-3 Cells

目的 探究姜黄素联合二甲双胍(Met)对卵巢癌细胞SKOV-3凋亡的影响。方法 将培养的卵巢癌细胞SKOV-3分成对照组、姜黄素组、二甲双胍组、联用组,采用CCK-8实验检测对细胞增殖抑制的影响,利用Annexin V-FITC/PI染色法检测对SKOV-3细胞凋亡的影响。结果 SKOV-3给药48 h后,CCK-8结果显示单药组呈时间、浓度依赖性地抑制细胞生长,联用组更能抑制细胞生长。Annexin V-FITC/PI染色法检测发现给药浓度越高,细胞凋亡率越高,且联用组促进凋亡能力比单药组较强。结论 Met、姜黄素呈浓度依赖性地抑制SKOV-3细胞生长,姜黄素和Met联用比单独使用抑制细胞增殖,促进凋亡的效果更好。

OBJECTIVE To investigate the effect of curcumin combined with metformin(Met)on SKOV-3 apoptosis of ovarian cancer cells.METHODS The cultured ovarian cancer cell line SKOV-3 was divided into control group,curcumin group,metformin group and combination group.The effect on cell proliferation inhibition was detected by CCK-8 assay and apoptosis was detected by Annexin V-FITC/PI staining.RESULTS 48 h after SKOV-3 administration,CCK-8 results showed that the cell growth was time and concentration dependent in the monotherapy group,and the cell growth was more inhibited in the combination group.Annexin V-FITC/PI staining showed that the higher the concentration,the higher the apoptosis rate,and the combination group was stronger than the monotherapy group in promoting apoptosis.CONCLUSION Metformin and curcumin inhibit the growth of SKOV-3 cells in a concentration-dependent manner.Curcumin and metformin in combination has a better effect on inhibiting cell proliferation and promoting apoptosis than alone.
