基于宏基因组的储粮害虫嗜卷书虱共生微生物多样性及功能分析

Analysis on Diversity, Functions of Symbiotic Microorganisms of Liposcelisbostrychophila (Psocoptera: Liposcelididae) in Stored Grain Based on Metagenome

摘要 孤雌生殖的嗜卷书虱Liposcelis bostrychophila是储粮生态系统中常见害虫的优势种群之一,其共生微生物影响着嗜卷书虱L.bostrychophila的生物学特性。本研究采用宏基因组测序技术和生物信息技术,对HBSJ品系6000头试虫菌群DNA样本进行了测序,分析了嗜卷书虱L.bostrychophila共生微生物的多样性及功能。结果表明:共鉴定出10门51属146种;变形菌门和厚壁菌门为菌群的优势门,平均相对丰度分别为90.05%和9.21%;立克次氏体属和葡萄球菌属为菌群的优势属,平均相对丰度分别为88.99%和9.03%。猫立克次氏体和木糖葡萄球菌为菌群的优势种,平均相对丰度分别为70.51%和4.70%。嗜卷书虱L.bostrychophila成虫体内微生物参与的碳水化合物代谢通路和复制与修复通路的丰度最高,平均相对丰度分别为9.0%和6.8%,表明嗜卷书虱L.bostrychophila体内共生微生物在宿主取食后协助碳水化合物的代谢,以及参与宿主部分DNA的复制和损失修复。 Parthenogenetic Liposcelis bostrychophila is one of the dominant pest populations easily occurring in the grain storage ecosystem,and its symbiotic microorganisms affect the biological characteristics of the L.bostrychophila.In this paper,based on metagenomic sequencing technology and bioinformation technology,DNA samples of L.bostrychophila HBSJ of 6000 individuals were sequenced,and the diversity and function of the symbiotic microorganisms of L.bostrychophila were analyzed.The results indicated that:146 species symbiotic microorganisms of L.bostrychophila were identified from 51 genera of 10 phyla.Proteobacteria and Firmicutes were the dominant phyla,with an average relative abundance of 90.05%and 9.21%,respectively.Rickettsia and Staphylococcus were the dominant genera,with average relative abundance of 88.99%and 9.03%,respectively.Rickettsia_felis and Staphylococcus_xylosus were the dominant species,with an average relative abundance of 70.05%and 4.70%,respectively.The abundance related to carbohydrate metabolism pathway and replication and repair pathway in L.bostrychophila adult were high,with an average relative abundance of 9.0%and 6.8%,respectively.It indicated that the symbiotic microorganisms in bookworm assisted the metabolism of carbohydrate and participated in the replication and repair of partial DNA of the host after feeding.
