摘要
为研发一种以桦褐孔菌和山楂为原料药制备的咀嚼片,并对成品的降血脂作用进行验证。本研究采用单因素方法和响应面试验优化咀嚼片最优制备工艺,测定小鼠血清中胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)、谷丙转氨酶(ALT)及谷草转氨酶(AST)的水平变化,判断咀嚼片降血脂作用。结果表明,最佳处方工艺为30%主药,42.48%淀粉,26%甘露醇,0.02%阿斯巴甜以及1.5%硬脂酸镁;60%乙醇为润湿剂,过14目筛制粒,65℃干燥35 min。制备所得的咀嚼片可显著降低TC、TG、LDL-C、ALT、AST水平,复方桦褐孔菌咀嚼片可以显著改善高脂血症小鼠血脂水平,修复高脂饮食诱发的肝细胞损伤,具有明显的降血脂作用。
To develop a chewing tablet with Inonotus obliquus and hawthorn as raw materials,and verify the effect of the finished product on reducing blood lipid.Single factor method and response surface test in this study were used to optimize the preparation process of the chewable tablets.The effect of the chewable tablets on reducing blood lipid was determined by determining the levels of cholesterol(TC),triglyceride(TG),low-density lipoprotein(LDL-C),high-density lipoprotein(HDL-C),alanine aminotransferase(ALT) and aspartate aminotransferase(AST) in the serum of mice.The optimum formulation was 30% main drug,42.48% starch,26% mannitol,0.02% aspartame and 1.5% magnesium stearate;the wet granulation method was selected,with 60% ethanol as the wetting agent,passing through 14 mesh sieve for granulation,and drying at 65℃ for 35 min.The prepared chewable tablets can significantly reduce the levels of TC,TG,LDL-C,ALT,AST;compound Inonotus obliquus chewable tablets can significantlyimprove the blood lipid level of hyperlipidemia mice,repair the damage of myocardial cells and hepatocytes induced by high-fat diet,and have a significant role in reducing blood lipid.
