基于NF-κB信号通路分析电针对腰椎间盘突出症大鼠模型炎症反应的影响

摘要 目的:探讨电针对腰椎间盘突出症大鼠模型炎症反应的影响及机制。方法:选取40只SD大鼠作为研究对象,将其随机分为正常对照组、模型组、假手术组及电针组,每组各10只。分别在造模前1 d、造模后3 d和7 d检测各组大鼠热痛刺激缩爪阈值;采用HE染色观察腰脊神经根形态学变化;采用ELISA方法检测大鼠血清中TNF-α、IL-1β和IL-6的含量;采用WB及PCR检测大鼠脊髓中IKK-β、NF-κB p65、IkBa蛋白及mRNA表达。结果:与造模前比较,造模后3 d模型组和电针组热痛刺激缩爪阈值降低,符合腰椎间盘突出症对痛觉敏感的特点,造模成功,造模后7 d电针组阈值有所升高(P<0.05);电针可改善腰脊神经根形态学变化;与对照组比较,模型组、电针组TNF-α、IL-1β和IL-6表达较高,与模型组比较,电针组TNF-α、IL-1β和IL-6表达较低(P<0.05);与对照组比较,模型组、电针组IKK-β、NF-κB p65及IkBa蛋白表达较高,与模型组比较,电针组IKK-β、NF-κB p65及IkBa蛋白表达较低(P<0.05);与对照组比较,模型组、电针组IKK-β、NF-κB p65及IkBa mRNA表达较高,与模型组比较,电针组IKK-β、NF-κB p65和IkBa mRNA表达较低(P<0.05)。结论:电针可以改善腰椎间盘突出症大鼠炎症反应,可能是通过调控NF-κB实现的。 Objective:To investigate effect and mechanism of electro-acupuncture(EA)on inflammatory response in rat model of lumbar disc herniation(LDH).Methods:40 SD rats were randomly divided into the control group,the model group,the sham operation group and the EA group,with 10 rats in each group.The threshold of thermal pain stimulation claw withdraw was recorded at 1 d before modeling,3 d and 7 d after modeling.The morphological changes of the lumbar spinal nerve roots were observed by HE staining;the serum levels of TNF-α,IL-1βand IL-6 were measured by ELISA;the protein and mRNA expressions of IKK-β,NF-κB p65 and IkBa in the spinal cord were detected by WB and PCR,respectively.Results:Compared with that in the pre-modeling period,the threshold of thermal pain stimulation claw withdraw was lower in the model group and the EA group 3 days after modeling,which was consistent with the characteristics of pain sensitivity in LDH,showing successful modeling;the threshold was increased in the EA group 7 days after modeling(P<0.05).EA could improve the morphological changes of lumbar spinal nerve root.Compared with those in the control group,the expressions of TNF-α,IL-1βand IL-6 were higher in the model group and the EA group;compared with those in the model group,the expressions of TNF-α,IL-1βand IL-6 were lower in the EA group(P<0.05).Compared with those in the control group,the protein expressions of IKK-β,NF-κB p65 and IkBa were higher in the model group and the EA group;compared with those in the model group,the protein expressions of IKK-β,NF-κB p65 and IkBa were lower in the EA group(P<0.05).Compared with those in the control group,the mRNA expressions of IKK-β,NF-κB p65 and IkBa were higher in the model group and the EA group(P<0.05);compared with those in the model group,the mRNA expressions of IKK-β,NF-κB p65 and IkBa were lower in the EA group(P<0.05).Conclusion:EA can improve inflammatory response in rats with LDH,which is possibly achieved by modulating NF-κB.
