摘要
目的:探讨湖北地区儿童VDR基因(rs2239185、rs7975232、rs2525046、rs2228570)和GSDMB基因(rs2305480)单核苷酸多态性(SNP)与儿童哮喘的易感性。方法:采用前瞻性研究方法,纳入158例哮喘患儿(哮喘组)和156例健康儿童(对照组),运用Sequenom MassArray质谱芯片微列阵技术对两组对象VDR和GSDMB基因的多个SNP位点进行基因分型,分析上述各位点在两组间分布的差异,筛选湖北地区儿童哮喘易感SNP位点。结果:GSDMB基因rs2305480位点GG、AG、AA 3种基因型频率在哮喘组中分别为64.56%、4.43%、31.01%,在对照组中分别为51.28%、8.97%、39.74%,两组间GG基因型分布频率差异具有统计学意义(P<0.05);GSDMB基因rs2305480位点的等位基因G、A在哮喘组中的分布频率分别为80.06%、19.94%,在对照组中的分布频率分别为71.15%、28.85%,两组间的等位基因A分布频率差异具有统计学意义(P<0.05);GSDMB基因rs2305480位点不同基因分布哮喘患儿白细胞计数、中性粒细胞百分比、淋巴细胞百分比和嗜酸性粒细胞百分比的差异无统计学意义(P>0.05);两组间VDR基因rs2239185、rs7975232、rs2525046、rs2228570位点多态性分布差异无统计学意义(P>0.05)。结论:GSDMB基因rs2305480位点多态性与我国湖北地区儿童哮喘的易感性相关:VDR基因rs2239185、rs7975232、rs2525046、rs2228570位点多态性与我国湖北地区儿童哮喘易感性无显著相关性。
Objective:To investigate the potential relationship between polymorphisms of VDR gene(rs2239185,rs7975232,rs2525046,rs2228570),GSDMB gene(rs2305480)and bronchial asthma of children in hubei province.Method:158 asthmatic children(asthma group)and 156 healthy children(control group)were recruited in this study.The genotypes of multiple single nucleotide polymorphisms(SNPs)loci in VDR and GSDMB genes in the two groups were analyzed by Sequenom MassArray spectrometry chip microarray technology.The differences in the distribution of these SNPs between the two groups were analyzed,so as to screen for the susceptibility to asthmatic children in hubei province.Results:The frequencies of three genotypes(GG,AG and AA)in rs2305480 were 64.56%,4.43%and 31.01%in the asthma group,and 51.28%,8.97%and 39.74%in the control group,and the difference of GG genotypes between the two groups was statistically significant(P<0.05).In asthma group,the frequencies of the alleles G and A in rs2305480 were 80.06%,19.94%,and 71.15%,28.85%in the control group.The difference in gene frequencies of allele A between the two groups was statistically significant(P<0.05).Among asthmatic children with different genotype of rs2305480 in gene GSDMB,the WBC,neutrophils(NEU%),lymphocytecounts(LYM%)and ensinophils(EOS%)were not significantly different(P>0.05).There was no significant difference in the distribution of rs2239185,rs7975232,rs2525046,rs2228570 in geng VDR between the asthma grounp and the healthy control grounp.Conclusion:The polymorphism of rs2305480 in GSDMB gene was associated with the susceptibility to childhood asthma in hubei province,while the SNPs of rs2239185,rs7975232,rs2525046,rs2228570 in geng VDR have no signficant correlation with childhood asthma in hubei province.
