基于基因表达综合数据库筛选急性冠脉综合征的差异基因和生物信息学分析

摘要 目的对急性冠脉综合征(ACS)患者和正常患者的mRNA基因芯片数据进行生物信息学分析,筛选ACS差异表达基因,确定ACS的核心基因。方法利用基因表达综合数据库(GEO)下载的数据筛选ACS患者与正常患者之间差异表达基因,对差异表达基因进行基因功能(GO)富集分析,使用String在线数据库构建基因的蛋白互作网络(PPI),并确定ACS的核心基因。结果筛选出ACS患者与正常患者之间存在350个差异基因,上调基因244个,下调基因106个;GO富集分析结果显示,差异基因主要富集于核糖体相关的信号通路;构建差异表达基因的PPI网络共筛选出20个核心基因,治疗后ACS患者与正常患者基本转录因子3(BTF3)基因表达比较差异有统计学意义(P<0.05),两者间其他基因表达比较差异无统计学意义。结论筛选出的20个核心基因可作为ACS的核心基因,为ACS的治疗和研究提供理论指导。 Objective To analyze the mRNA microarray data of patients with acute coronary syndrome(ACS)and normal patients by bioinfor-matics analysis,screen the differentially expressed genes in ACS,and determine the core genes of ACS.Methods The data downloaded from the gene expression omnibus(GEO)were used to screen the differentially expressed genes between ACS patients and normal patients,and gene ontology(GO)enrichment analysis was implemented on the differentially expressed genes,protein-protein interaction network(PPI)of genes was structured by String online database,and the core genes of ACS were determined.Results There were 350 differential genes between ACS patients and normal patients,244 up-regulated genes and 106 down-regulated genes;GO enrichment analysis showed that the differential genes were mainly concentrated in ribosomal related signaling pathways.A total of 20 core genes were screened out by constructing the PPI network of differentially expressed genes,and there were statistically significant differences in basic transcription factor 3(BTF3)gene expression between ACS patients after treatment and normal patients(P<0.05),while there were no significant differences in other core genes expression between ACS patients after treatment and nor-mal patients.Conclusion The selected 20 core genes can be used as the core genes of ACS,providing theoretical guidance for the treatment and re-search of ACS.
