摘要
目的 探讨亮菌口服液中亮菌多糖对结肠癌、黑色素瘤和肉瘤等恶性肿瘤生长、增殖和侵袭的影响。方法 离体水平分别对人结肠癌细胞系HCT116细胞和鼠转移性黑色素瘤细胞系B16-F10细胞加入不同浓度(10μg·m L^(-1)、100μg·m L^(-1)和250μg·m L^(-1))的亮菌多糖(亮菌口服液中提取),采用MTT法检测亮菌多糖对结肠癌、黑色素瘤的细胞毒性作用。在体水平选取BALB/c雌性小鼠24只,分别随机设置肉瘤荷瘤对照组(将浓度为2×10^(6)cells/500μL的肉瘤腹水肿瘤细胞,经皮下植入小鼠左腋窝)、低剂量亮菌多糖干预组(腹腔注射25 mg·kg^(-1))和高剂量亮菌多糖干预组(腹腔注射50 mg·kg^(-1)),分析不同浓度亮菌多糖对荷瘤小鼠外周血生化指标及肝、肾组织病理形态的影响。结果 10μg·m L^(-1)、100μg·mL^(-1)和250μg·mL^(-1)硫酸化亮菌多糖未能显著抑制上述肿瘤细胞的生长(P>0.05);25 mg·kg^(-1)和50 mg·kg^(-1)的亮菌多糖对肿瘤生长抑制均强于对照组,诱导荷瘤小鼠脾脏重量及外周血的血小板计数、AST水平增加均高于对照组(P<0.05)。结论 在体水平下,亮菌多糖可延缓肉瘤在荷瘤小鼠体内的生长和增殖,且对荷瘤小鼠没有显著的肝肾毒性,但对结肠癌、黑色素瘤的生长的影响不大。
Objective To explore the effects of armillariella polysaccharide in armillariella oral solution on the growth,proliferation and invasion of malignant tumors such as colon cancer,melanoma and sarcoma.Methods In vitro,different concentrations(10,100 and 250μg·mL^(-1))of armillariella polysacharide(extracted from armillariella oral solution)were added to human colon cancer cell line HCT116 cells and mouse metastatic melanoma cell line B16-F10 cells.MTT assay was used to detect the cytotoxic effect of armillariella polysaccharide on colon cancer and melanoma.24 BALB/c female mice were selected at the in vivo level and randomly divided into the sarcoma-bearing control group(sarcoma ascites tumor cells with 2x10^(6)cells/500μL were subcutaneously implanted into the left axilla of mice),the low-dose armillariella polysaccharide intervention group(intraperitoneal injection of 25 mg·kg'armillariella polysaccharide)and the high-dose armillariella polysaccharide intervention group(intraperitoneal injection of 50 mg·kg'armillariella polysaccharide).The effects of different concentrations of armillariella polysaccharide on peripheral blood biochemical indicators and pathological morphology of liver-kidney tissues in tumor-bearing mice were analyzed.Results 10,100 and 250μg·mL^(-1)of sulfated armillariella polysaccharide could not significantly inhibit the growth of the above tumor cells(P>0.05).In the low-dose armillariella polysaccharide intervention group and the high-dose armillariella polysaccharide intervention group,the inhibitory effects on tumor growth were stronger than those in the sarcoma-bearing control group,and the induction of increased spleen weight,peripheral blood platelet count and AST levels in tumor-bearing mice was higher than that in the sarcoma-bearing control group(P<0.05).Conclusion At the in vivo level,armillariella polysaccharide can delay the growth and proliferation of sarcoma in tumor-bearing mice,and it has no significant hepatorenal toxicity on tumor-bearing mice,and has little effect on the growth of colon cancer and melanoma.
