高效液相色谱法测定面包、糕点及饼干中脱氢乙酸及其钠盐的含量

摘要 目的:建立快速、简便地检测面包、糕点及饼干中脱氢乙酸及其钠盐含量的高效液相色谱法。方法:参照并优化《食品安全国家标准食品中脱氢乙酸的测定》(GB 5009.121—2016)中的前处理方法,采用C18色谱柱对样品进行分离,紫外检测器进行检测。结果:脱氢乙酸在1.0~200.0μg·mL^(-1)质量浓度范围内与峰面积呈良好的线性关系,相关系数为1.000 0;方法检出限为0.001 g·kg^(-1),定量限为0.003 g·kg^(-1);在3个加标水平下,回收率为97.3%~102.1%,RSD为0.21%~1.42%,均满足标准要求。结论:该方法具有快速、简便、准确度高等特点,适用于面包、糕点及饼干中脱氢乙酸及其钠盐含量的检测。 Objective:To establish a high performance liquid chromatography(HPLC) method for the rapid and simple determination of dehydroacetic acid and its sodium salt in bread,pastries and cookies.Method:The pretreatment method in GB 5009.121—2016 was referred to and optimized,and the samples were separated on a C_(18) column and detected by an ultraviolet detector.Result:Dehydroacetic acid showed a good linear relationship with the peak area in the mass concentration range of 1.0 ~ 200.0 μg·mL~(-1) with a correlation coefficient of 1.000 0.The limit of detection of the method was 0.001 g·kg~(-1),and the limit of quantification was 0.003 g·kg~(-1).The recoveries ranged of 97.3% ~ 102.1% with RSD of 0.21% ~ 1.42% at three spiked levels,all meeting the standard requirements.Conclusion:The method is rapid,simple,accurate and suitable for the determination of dehydroacetic acid and its sodium salt in bread,pastries and cookies.
