环状RNA HIPK3在急性胰腺炎患者中的表达及与病情严重程度和预后的相关性分析

Expression of Circular RNA HIPK3 in Patients with Acute Pancreatitis and its Correlation with Disease Severity and Prognosis

目的探讨环状RNA同源域相互作用蛋白激酶3(circHIPK3)在急性胰腺炎(AP)患者中的表达及与病情严重程度和预后的相关性。方法选取2020年11月至2021年11月我院126例AP患者为AP组,其中轻度38例,中度43例,重度45例;90例正常健康体检者为对照组。根据AP患者入院后28 d的生存情况分为生存组(n=86)和死亡组(n=40)。采用实时荧光定量PCR法(qRT-PCR)检测circHIPK3的表达水平。Pearson法分析circHIPK3表达水平与APACHEⅡ评分及Ranson评分的相关性;Logistic回归分析影响AP患者预后的危险因素。结果AP组的circHIPK3、IL-6、IL-8、TNF-α表达水平显著高于对照组(P<0.05);重度组circHIPK3、IL-6、IL-8、TNF-α的表达水平、APACHEⅡ评分及Ranson评分显著高于中度组及轻度组,中度组显著高于轻度组(P<0.05);circHIPK3的表达水平与IL-6、IL-8、TNF-α水平、APACHEⅡ评分及Ranson评分均呈正相关(P<0.05);生存组的Cr、CRP、cTn1、CK、BNP、circHIPK3水平均明显低于死亡组(P<0.05);Logistic回归分析发现CK、BNP、circHIPK3高水平是影响AP患者预后的危险因素(P<0.05)。结论AP患者血清circHIPK3表达水平异常增加,其水平与患者预后密切相关。

Objective To investigate the expression of circular RNA homeodomain-interacting protein kinase 3(circHIPK3)in patients with acute pancreatitis(AP)and its correlation with disease severity and prognosis.Methods From November 2020 to November 2021,126 AP patients in our hospital were regarded as AP group,including 38 mild cases,43 moderate cases,and 45 severe cases.90 normal healthy subjects were the control group.AP patients were grouped into survival group(n=86)and death group(n=40)according to the survival status of AP patients 28 days after admission.Real-time quantitative PCR(qRT-PCR)was used to detect the expression level of circHIPK3,Pearson method was used to analyze the correlation between the expression level of circHIPK3,APACHE II score and Ranson score;Logistic regression was used to analyze the risk factors affecting the prognosis of AP patients.Results The expression level of circHIPK3,IL-6,IL-8 and TNF-αin AP group was significantly higher than that in control group(P<0.05);The expression level of circHIPK3,IL-6,IL-8,TNF-α,APACHE II score and Ranson score in severe group was significantly higher than that in moderate and mild groups,and the moderate group was significantly higher than that in mild group(P<0.05);circHIPK3 expression level was positively correlated with IL-6,IL-8,TNF-αlevel,APACHE II score and Ranson score(P<0.05);the levels of Cr,CRP,cTn1,CK,BNP and circHIPK3 in the survival group were greatly lower than those in the death group(P<0.05);Logistic regression analysis showed that high levels of CK,BNP and circHIPK3 were risk factors affecting the prognosis of AP patients(P<0.05).Conclusion The expression level of serum circHIPK3 in AP patients is abnormally increased,and its level is closely related to the prognosis of patients.
