吉西他滨诱导的胰腺癌细胞外泌体促进胰腺癌细胞的增殖和侵袭

摘要 目的探讨正常培养和吉西他滨处理后胰腺癌细胞分泌的外泌体对胰腺癌细胞增殖和侵袭的作用。方法收集正常培养和吉西他滨处理后胰腺癌细胞上清液中的外泌体,利用透射电镜、纳米颗粒跟踪分析和Western blot法鉴定外泌体,分析外泌体的浓度和大小,将外泌体与胰腺癌细胞共孵育后检测细胞的增殖和侵袭能力,Western blot法检测CD147分子表达水平。结果提取到的外泌体表达外泌体特异性的标志物,吉西他滨处理后外泌体的浓度和大小没有显著变化,与外泌体共孵育后胰腺癌细胞的增殖和侵袭显著增强,而与吉西他滨诱导的胰腺癌细胞外泌体共孵育后细胞的增殖和侵袭进一步增强,CD147表达水平明显升高。结论吉西他滨诱导的胰腺癌细胞外泌体能够促进胰腺癌细胞的增殖和侵袭。 Objective To explore the effects of exosomes derived from pancreatic cancer cells treated with or without gemcitabine on the proliferation and invasion of pancreatic cancer cells.Methods Exosomes were extracted from supernatant of pancreatic cancer cells treated with or without gemcitabine.Transmission electron microscopy,nanoparticle tracking analysis and Western blot method were used to identify exosomes and the concentration and size of exosomes were determined.Incubation of exosomes with pancreatic cancer cells was performed to detect the proliferation and invasion of pancreatic cancer cells,and Western blot method was used to detect the expression of CD147.Results Exosome-specific markers were highly expressed in extracted exosomes.There was no significant difference of concentration and size of exosomes derived from pancreatic cancer cells treated with or without gemcitabine.Incubation with exosomes derived from pancreatic cancer cells promoted the proliferation and invasion of pancreatic cancer cells,and incubation with exosomes derived from pancreatic cancer cells treated with gemcitabine promoted the proliferation and invasion further,and the expression level of CD147 was upregulated significantly in gemcitabine-induced exosomes.Conclusion Gemcitabine-induced exosomes derived from pancreatic cancer cells can promote the proliferation and invasion of pancreatic cancer cells.
