榴莲酚酸类成分的制备及其对帕金森细胞模型活性影响的时程变化

Preparation of Phenolic Acid Extract from Durian Pulp and its Effect on Parkinson's Disease Cell Model

摘要 本研究旨在探讨不同浓度剂量榴莲酚酸类成分对帕金森细胞模型在不同时间点细胞活性的影响,以期了解其抗氧化的作用。采用当季新鲜成熟榴莲果肉,经过冻干研磨,多种有机溶剂提取、浓缩、冻干,制备榴莲酚酸类提取物,采用液质联用分析榴莲提取物成分。将神经母细胞瘤细胞接种于96孔板,分为:正常组、模型对照组、治疗组。在造模成功24 h后,对治疗组加入不同剂量榴莲酚酸类提取物,分别于24、36、48 h后,用CCK8试剂盒进行检测,观察榴莲提取物作用于帕金森细胞模型不同时间点的细胞活性。榴莲果肉中含有20余种化学成分,以酚酸类物质偏多,其中3,4-二羟基扁桃酸含量最高。加入不同浓度的榴莲酚酸类提取物后与正常组相比,24、36、48 h组细胞活性较模型组均有明显的升高,并且36、48 h中部分组间有明显的量效关系。本研究表明,榴莲果肉在提取的过程中因为光、温度等因素的影响多酚类物质可能会转变成酚酸类成分,这些酚酸类成分仍具有促进细胞活性,抗氧化的作用。不同浓度剂量榴莲酚酸类提取物对帕金森病细胞模型不同时间点细胞活性具有明显的促进作用,并随着时间的增加,促进作用更加明显。 This study aims to explore the effects of different concentrations and doses of phenolic acid extract from durian pulp on the cellular activity of Parkinson's cell models at different time points,and understand their antioxidant effects.The fresh and ripe durian pulp in season was used,and then it was freeze-dried and ground,extracted,concentrated and freeze-dried with various organic liquid-liquid extraction to prepare the durian extract.The electrospray ionization(ESI)positive ion mode was used to analyze the components of durian extract by LC-MS.Neuroblastoma cells were seeded into 96 well plates and divided into normal group,model control group and treatment group.After 24 hours of successful modeling,different doses of durian phenolic acid extracts were added to the treatment group.After 24 hours,36 hours,and 48 hours,the activity of durian extract on Parkinson's cell model cells at diffe-rent time points was observed using CCK8 reagent kit.The pulp extract of durian contains more than 20 chemical components,among which phenolic acids are abundant,and the content of 3,4-dihydroxyneneneba mandelic acid is the highest.After adding different concentrations of durian phenolic acid extract,compared with the normal group,the cell activity of the 24,36,and 48 hour groups was significantly increased compared to the model group,and there was a significant dose-effect relationship between some groups at 36 and 48 hours.This study indicates that during the extraction process of durian fruit flesh,polyphenols may be converted into phenolic acids due to factors such as light and temperature,which still have the effects of promoting cell activity and antioxidant activity.Different concentrations and doses of durian phenolic acid extract have a significant promoting effect on the cell activity of Parkinson's disease cell models at different time points,and the promoting effect becomes more significant with time.
