‘贵妃’杧果胚败育果实植物激素生物合成的转录组分析

摘要 【目的】为创制胚败育特异种质和优质胚败育杧果生产提供参考。【方法】以花后第14、16、18、20周4个发育时期的‘贵妃’杧果的胚正常果和胚败育果作为研究对象,基于转录组测序数据,探讨胚正常和胚败育杧果中乙烯、生长素、赤霉素、脱落酸、油菜素甾醇和茉莉酸6种植物激素生物合成途径中差异表达基因的表达趋势,研究其在胚正常和胚败育杧果中的作用和差异。选取15个具有指示和标记作用的关键差异表达基因进行荧光实时定量PCR,对转录组测序和分析结果的可靠性进行验证。【结果】‘贵妃’杧果胚正常和胚败育果实中参与植物激素生物合成途径的关键基因表达情况在不同发育阶段存在明显差异,且呈现一定规律。胚败育果中调控乙烯合成的关键基因在花后第14、16周的表达量高于胚正常果,而调控生长素合成的关键基因表达量低于胚正常果;调控茉莉酸合成的关键基因大多在胚败育果中表达量更高。荧光实时定量PCR的结果与转录组分析结果基本一致,表明结果真实可靠。【结论】乙烯、生长素和脱落酸共同调控了‘贵妃’杧果胚败育果的早熟,赤霉素调控了胚正常果果形更大,杧果在受到外界环境胁迫的时候,更容易产生胚败育果。 【Objective】To provide a reference for the creation of embryo-aborted germplasm and high-quality embryo-aborted mango production.【Method】The study used the transcriptome sequencing data to explore the expression pattern of differentially expressed genes(DEGs)involved in the biosynthesis pathway of six phytohormones which were ethylene,indole-3-acetate,gibberellin,abscisic acid,brassinosteroid and jasmonate in normal and embryo-aborted fruits to elucidate their roles and differences.Normal and embryo-aborted fruits of‘Guifei’mango at 14th,16th,18th,and 20th week after flowering was used in the study.In order to verify the reliability of transcriptome sequencing and analysis results,the experimental results were verified by real-time quantitative PCR.【Result】A total of sixty two differentially expressed genes were screened in the biosynthetic pathways of 6 phytohormone.The expression of the key genes involved in phytohormone biosynthesis pathways in normal and embryo-aborted mango was significantly different at various developmental stages and showed certain regularity.The expression of key genes which regulating ethylene synthesis in embryo-aborted fruits was higher than that in normal fruits at 14 and 16 weeks after flowering,while the expression of key genes which regulating auxin synthesis was lower than that in normal fruits.Most of the key genes regulating jasmonic acid synthesis were more highly expressed in embryo-aborted fruits.The results of real-time quantitative PCR were basically consistent with the results of transcriptome analysis which indicating that the results were true and reliable.【Conclusion】Ethylene,auxin and abscisic acid jointly regulated the early maturity of‘Guifei’mango fruit with aborted embryo.Gibberellin regulated the larger shape of normal mango.Mango was more likely to produce embryo-aborted fruit when subjected to external environmental stress.
