酶标仪法测定发酵液中谷胱甘肽的含量

摘要 在菌种选育阶段,为满足谷胱甘肽测定效率高、速度快的需求,本研究以酶标仪法测定了发酵液中谷胱甘肽的含量,并以标准曲线的线性回归方程计算谷胱甘肽含量,并对其进行统计,最终利用origin8.0软件将酶标仪与高效液相色谱的测定值进行拟合。结果显示,酶标仪法与高效液相色谱法在测定效果上具有较强的相似性,较高效液相色谱而言,酶标仪具有高效、快速及简单的测量优势。 In the stage of strain selection,in order to meet the requirements of high efficiency and fast detection of glutathione,this study used an enzyme-linked immunosorbent assay(ELISA)to determine the content of glutathione in the fermentation broth.The linear regression equation of the standard curve was used to calculate the glutathione content,and it was statistically analyzed.Finally,Origin8.0 software was used to fit the measured values of the ELISA with those of highperformance liquid chromatography.The results showed that the enzyme-linked immunosorbent assay(ELISA)method and high-performance liquid chromatography(HPLC)method have strong similarities in measurement effectiveness.Compared with HPLC,ELISA has the advantages of high efficiency,fast and simple measurement.
