不同病理类型及分期甲状腺癌组织中miR-197、miR-221和miR-222的表达及其临床意义

Expression of miR-197,miR-221 and miR-222 in thyroid cancer tissues of different pathological types and stages and their clinical significance

目的:观察不同病理类型及分期甲状腺癌组织中miR-197、miR-221和miR-222的表达,并分析其在甲状腺癌诊断中的意义。方法:回顾性分析2020年01月至2022年12月我院92例甲状腺癌患者临床资料,患者均进行甲状腺癌组织、癌旁组织病理检查,采用定量逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction, RT-PCR)测定miR-197、miR-221和miR-222表达水平,对比不同病理类型、分期等病理特征的甲状腺癌患者miR-197、miR-221和miR-222表达状况,并分析其在甲状腺癌诊断中的意义。结果:甲状腺癌组织中miR-197、miR-221、miR-222表达高于癌旁组织(t=14.696、14.004、15.351,P<0.05);淋巴结转移、未分化癌、Ⅲ-Ⅳ期的甲状腺癌患者miR-197、miR-221、miR-222表达分别高于未发生淋巴结转移、乳头状癌/髓样癌/滤泡癌、Ⅰ-Ⅱ期甲状腺癌患者(P<0.05)。经Logistic回归分析,结果显示,miR-197、miR-221、miR-222高表达是甲状腺癌患者发生淋巴结转移、发生未分化癌及是Ⅲ-Ⅳ期甲状腺癌的危险因素(OR>1,P<0.05)。结论:不同病理类型及分期甲状腺癌组织中miR-197、miR-221和miR-222的表达存在差异,三者将有助于判断甲状腺癌是否淋巴结转移、不同病理类型及临床分期。

Objective:To observe the expression of miR-197,miR-221 and miR-222 in different pathological types and stages of thyroid cancer,and analyze their significance in the diagnosis of thyroid cancer.Methods:The clinical data of 92 patients with thyroid cancer in our hospital from January 2020 to December 2022 were retrospectively analyzed.All patients underwent pathological examination of thyroid cancer tissues and adjacent tissues,and the expression levels of miR-197,miR-221 and miR-222 were determined by quantitative reverse transcription-polymerase chain reaction(RT-PCR),and the miR-197,miR-222 in patients with different pathological types and stages were compared.Results:The expressions of miR-197,miR-221 and miR-222 in thyroid cancer tissues were higher than those in adjacent tissues(t=14.696,14.004 and 15.351,P<0.05).The expressions of miR-197,miR-221 and miR-222 in patients with lymph node metastasis,undifferentiated carcinoma and stageⅢ-Ⅳthyroid cancer were higher than those in patients without lymph node metastasis,papillary carcinoma/medullary carcinoma/follicular carcinoma and stageⅠ-Ⅱthyroid cancer,respectively(P<0.05).Logistic regression analysis showed that the high expression of miR-197,miR-221 and miR-222 was a risk factor for lymph node metastasis,undifferentiated cancer and stageⅢ-Ⅳthyroid cancer(OR>1,P<0.05).Conclusion:The expressions of miR-197,miR-221 and miR-222 are different in different pathological types and stages of thyroid cancer,which will help to judge whether thyroid cancer has lymph node metastasis,different pathological types and clinical stages.
