基于“肾主水”理论的七叶胆总苷治疗膀胱癌大鼠的作用研究

摘要 目的基于“肾主水”理论观察膀胱灌注七叶胆总苷治疗N-甲基-N-亚硝基脲(MNU)诱导的SD大鼠膀胱癌(BC)的作用。方法取106只SPF级SD大鼠,随机分为空白组、模型组、低剂量组、中剂量组、高剂量组及阳性对照组,模型组、低剂量组、中剂量组、高剂量组及阳性对照组通过膀胱灌注的方式给予MNU进行造模,空白组和模型组予生理盐水0.5 ml膀胱灌注;低剂量组(20 mg/kg)、中剂量组(40 mg/kg)、高剂量组(80 mg/kg)予七叶胆药粉灌注给药;阳性对照组予卡介苗冻干粉灌注给药。通过组织学病理切片观察膀胱内的形态、计算各组膀胱脏器指数及检测相关生理指标,评价膀胱灌注七叶胆对MNU诱导SD大鼠BC的作用效果。结果第16周,模型组大鼠膀胱的脏器指标高于空白组,差异有统计学意义(P<0.05)。低剂量组、中剂量组、高剂量组和阳性对照组的大鼠膀胱脏器指标低于模型组,差异有统计学意义(P<0.05)。结论七叶胆总苷具有一定的抗膀胱癌的药理活性,可为抗膀胱癌药物开发提供一定的参考。 Objective To observe the effect of Gynostemma pentaphyllum on N-methyl-n-nitrosourea(MNU)-induced bladder cancer(BC)in SD rats based on the theory of"renal main water".Methods A total of 106 SPF SD rats were randomly divided into blank group,model group,low-dose group,medium-dose group,high-dose group and positive control group.Model group,low-dose group,medium-dose group,high-dose group and positive control group were given MNU by bladder perfusion for modeling,and blank group and model group were given 0.5 ml bladder perfusion of normal saline.Low dose group(20 mg/kg),medium dose group(40 mg/kg)and high dose group(80 mg/kg)were injected with aesculus chinensis powder.The positive control group was injected with lyophilized BCG vaccine powder.The morphology of the bladder in each group was observed by histological and pathological sections,and the bladder organ index of each group was calculated and related physiological indexes were detected.To evaluate the effect of bladder infusion of aesculus on BC induced by MNU in SD rats.Results At the 16th week,the bladder organ index of the model group was higher than that of the blank group,and the difference was statistically significant(P<0.05).The bladder organ indexes of low-dose group,medium-dose group,high-dose group and positive control group were lower than those of model group,and the differences were statistically significant(P<0.05).Conclusion Gynostemma pentaphyllum has certain pharmacological activity against bladder cancer.The experimental data can provide a certain reference for the development of anti-bladder cancer drugs.
