妇科再造丸含药血清对子宫肌瘤细胞增殖和凋亡的影响

Effects of Fukezaizao Pills-containing Serum on the Proliferation and Apoptosis of Uterine Fibroid Cells

目的:探讨妇科再造丸(FKW)含药血清对子宫肌瘤细胞增殖和凋亡的影响。方法:收集2020年1月—2022年6月于南方医科大学南方医院行肌瘤剔除术的32例子宫肌瘤患者的子宫肌瘤组织标本,对人子宫肌瘤原代细胞进行分离培养;将48只健康雌性SD大鼠随机分为无药血清组、低剂量组、中剂量组与高剂量组,各12只。无药血清组每次灌喂生理盐水10 mL/kg,低剂量组灌喂FKW 8.32 g/次,中剂量组灌喂FKW 16.9 g/次,高剂量组灌喂FKW 33.54 g/次,灌喂3 d,2次/d。取大鼠血清作用于培养的子宫肌瘤细胞,检测细胞的增殖活力、细胞凋亡率、蛋白酪氨酸磷酸酶基因(PTEN)与促细胞凋亡蛋白(Bax)表达情况。结果:各组吸光度值比较,无药血清组>低剂量组>中剂量组>高剂量组;各组细胞凋亡率比较,无药血清组<低剂量组<中剂量组<高剂量组;各组PTEN与Bax蛋白表达水平比较,无药血清组<低剂量组<中剂量组<高剂量组,差异有统计学意义(P<0.05)。结论:FKW含药血清对子宫肌瘤细胞增殖和凋亡具有一定的调控作用,可能通过上调PTEN与Bax蛋白表达抑制肌瘤生长。

Objective:To explore the effect of Fukezaizao Pills(FKW)-containing serum on the proliferation and apoptosis of uterine fibroid cells.Methods:Uterine fibroid tissue specimens from 32 patients with uterine fibroids who underwent myomectomy at Nanfang Hospital of Southern Medical University from January 2020 to June 2022 were collected,and human uterine fibroid primary cells were isolated and cultured.Forty-eight healthy female SD rats were randomly divided into drug-free serum group,low-dose group,medium-dose group and high-dose group,with 12 rats in each group.The drug-free serum group was fed with physiological saline 10 mL/kg each time,the low-dose group was fed with FKW 8.32 g/time,the middle-dose group was fed with FKW 16.9 g/time,and the high-dose group was fed with FKW 33.54 g/time,fed by gavage for 3 days,2 times/d.Rat serum was taken to act on cultured uterine fibroid cells to detect cell proliferation activity,cell apoptosis rate,protein tyrosine phosphatase gene(PTEN)and pro-apoptotic protein(Bax)expression.Results:Comparison of the absorbance values of each group,drug-free serum group>low-dose group>mid-dose group>high-dose group;comparison of cell apoptosis rate in each group,drug-free serum group<low-dose group<mid-dose group<high-dose group;Comparing the expression levels of PTEN and Bax proteins in each group,the drug-free serum group<low-dose group<mid-dose group<high-dose group,the difference was statistically significant(P<0.05).Conclusion:FKW-containing serum has a certain regulatory effect on uterine fibroid cell proliferation and apoptosis,and may inhibit fibroid growth by up-regulating the expression of PTEN and Bax proteins.
