摘要
为建立“大孔吸附树脂柱色谱-凝胶柱色谱-HPLC”三步法快速制备绶草中活性成分Spiranthol-A方法,并评价其抑制乙酰胆碱酯酶活性,在HPLC导向下,将绶草95%乙醇提取物先经大孔吸附树脂柱色谱分离得到含Spiranthol-A部位,再通过Sephadex-LH 20柱色谱和HPLC纯化,得单体化合物Spiranthol-A;采用微孔板法评价Spiranthol-A体外抑制乙酰胆碱酯酶活性。结果表明:Spiranthol-A纯度较高且抑制乙酰胆碱酯酶活性较强,IC_(50)值仅为(10.58±0.44)μg·mL^(-1)。综上,该研究所建立的快速制备Spiranthol-A方法具有一定的应用前景,且Spiranthol-A具有显著抑制乙酰胆碱酯酶作用,值得深入研究。
In order to establish a “macroporous adsorption resin column chromatography-gel column chromatography-HPLC” three-step rapid preparation method and evaluate the acetylcholinesterase(AChE)-inhibitory activity of Spiranthol-A, an active compound in Spiranthes sinensis, the 95% ethanol extract of S.sinensis was separated by macroporous adsorption resin column chromatography to obtain the fraction containing Spiranthol-A, which was then purified by the Sephadex-LH 20 column chromatography and HPLC to get Spiranthol-A, guided by the HPLC. In addition, the in vitro AChE-inhibitory activity of Spiranthol-A was tested by a microplate assay. The results showed that Spiranthol-A obtained showed a high purity and a strong AChE-inhibitory activity, with an IC_(50) value of(10.58±0.44) μg·mL^(-1). The rapid preparation method established in this paper has a certain application prospect. Spiranthol-A has a significant AChE-inhibitory effect and is worthy of a further study.
