HPK1 GROβ及THRSP蛋白在乳腺癌患者中的表达及与预后的相关性分析

Expressions of HPK1,GROβand THRSP proteins in breast cancer patients and their correlations with prognosis

摘要 目的 分析造血祖细胞激酶1(HPK1)、生长调节蛋白-β(GROβ)及THRSP蛋白在乳腺癌患者中的表达及与预后的相关性。方法 选取2016年1月—2019年12月温州市中医院收治的乳腺癌患者100例,取所有患者手术切除的癌组织及癌旁组织标本,采用免疫组化染色法检测GROβ水平,Western blot法检测HPK1、THRSP表达量。结果 癌旁组织中HPK1、GROβ及THRSP蛋白表达量分别为(1.62±0.35)、(47.23±6.56)pg/ml、(1.13±0.16);癌组织中HPK1、GROβ及THRSP蛋白表达量分别为(1.16±0.19)、(116.35±25.59)pg/ml、(1.57±0.31)。与癌旁组织相比,癌组织中HPK1表达量较低,GROβ、 THRSP蛋白表达量较高(均P<0.05)。淋巴结转移、Ⅲ期、Ⅳ期、低分化患者中HPK1表达量较低,GROβ、 THRSP蛋白表达量较高(均P<0.05)。乳腺癌癌组织中HPK1与GROβ表达量呈负相关(r=-0.292,P<0.05),GROβ与THRSP表达量呈正相关(r=0.277,P<0.05),HPK1与THRSP表达量呈负相关(r=-0.348,P<0.05)。多因素logistic回归分析结果显示:HPK1、 GROβ、 THRSP蛋白是乳腺癌患者预后的危险因素(均P<0.05)。结论 HPK1、GROβ及THRSP蛋白在乳腺癌组织中表达异常,与TNM分期、分化程度、有无淋巴结转移相关,三者表达和乳腺癌患者预后有一定相关性。 Objective To analyze the expressions of hematopoietic progenitor cell kinase 1(HPK1),growth regulator protein-β(GROβ),and THRSP proteins in breast cancer patients and their correlations with prognosis.Methods A total of 100 breast cancer patients were selected from Wenzhou Hospital of Traditional Chinese Medicine from January 2016 to December 2019,the breast cancer tissue samples and adjacent tissue samples after surgical resection were obtained,GROβlevels were detected by immunohistochemical staining method,HPK1 and THRSP expressions were detected by Western blot.Results The expression levels of HPK1,GROβ,and THRSP proteins in adjacent tissue of breast cancer were(1.62±0.35),(47.23±6.56)pg/ml,and(1.13±0.16),respectively.The expression levels of HPK1,GROβ,and THRSP proteins in breast cancer tissue were(1.16±0.19),(116.35±25.59)pg/ml,and(1.57±0.31),respectively.Compared with adjacent tissue of breast cancer,the expression level of HPK1 in breast cancer tissue was lower,the expression levels of GROβand THRSP proteins were higher(P<0.05).The expression levels of HPK1 in breast cancer patients of lymph node metastasis,stageⅢ,stageⅣ,and poor differentiation were lower,the expression levels of GROβand THRSP proteins were higher(P<0.05).In breast cancer tissue,HPK1 expression was negatively correlated with GROβexpression(r=-0.292,P<0.05),GROβexpression was positively correlated with THRSP expression(r=0.277,P<0.05),HPK1 expression was negatively correlated with THRSP expression(r=-0.348,P<0.05).Multivariate logistic regression analysis showed that HPK1,GROβ,and THRSP proteins were risk factors of prognosis of breast cancer patients(all P<0.05).Conclusion The expression levels of HPK1,GROβ,and THRSP proteins in breast cancer tissues are significantly abnormal,which are correlated with TNM stage,differentiation degree,and the presence or absence of lymph node metastasis,and there is a certain correlation between the expressions of the three proteins and the prognosis of breast cancer patients.
