新方颠倒散体外抑制痤疮丙酸杆菌实验研究及皮肤毒性测试

摘要 目的:研究不同配比新方颠倒散水提物溶液对痤疮丙酸杆菌体外的抑菌作用。方法:配制不同组分新方颠倒散水提物溶液A、C、E、G、I、K,颠倒散水提物溶液(M)为阳性对照,另设阴性对照、空白对照;采用37℃厌氧培养法对痤疮丙酸杆菌进行培养,观察各溶液体外的抑菌效果;并采用单次限量法,取A进行大鼠急性经皮毒性试验。结果:培养48 h后,空白对照无菌落,其余均见菌落,各新方颠倒散水提物溶液分别与阴性对照比较,均有显著统计学意义(P<0.05),与阳性对照比较均无显著统计学意义(P>0.05)。大鼠急性经皮毒性试验:A组、对照组、M组反应平均值分别为0、0、1.2,生理情况正常。结论:不同配比新方颠倒散水提物溶液体外对痤疮丙酸杆菌均有抑制作用,相互比较以及与颠倒散比较均无显著差异,且新方颠倒散溶液无皮肤刺激及中毒反应。 Objective:Study the in vitro antibacterial effect of different ratios of Xinfang Diandao powder aqueous extracts on cutibacterium acnes.Methods:The Xinfang Diandao powder water extract solutions A,C,E,G,I,and K of different components were prepared according to the ratio of sulfur,rhubarb,and saponin horn at 1∶1∶1,1∶4∶4,1∶8∶8,1∶12∶12,1∶16∶16,and 1∶20∶20,respectively.Diandao Powder aqueous extract solution(M)serves as a positive control,with additional negative and blank controls.Cutibacterium acnes was cultured under anaerobic condition at 37℃.Observe the antibacterial effect of each solution in vitro.And using the single dose limit method,A was taken for acute percutaneous toxicity test in rats.Results:After 48 hours of cultivation,the blank control showed no colonies,while the rest showed colonies.The water extract solution of each new formula showed significant statistical significance compared to the negative control(P<0.05),while there was no significant statistical significance compared to the positive control(P>0.05).Acute percutaneous toxicity test in rats:The average response values of Group A,Control Group,and Group M were 0,0,and 1.2,respectively.The physiological conditions of the rats in each group were normal.Conclusion:Different ratios of the new formula Xinfang Diandao powder aqueous extract solution have inhibitory effects on Propionibacterium acnes in vitro.There was no significant difference in comparison with each other or in comparison with Diandao powder aqueous extract solution.And the Xinfang Diandao water extract solution has no skin irritation or toxic reactions.
