基于UPLC-CAD的不同产地麦冬及不同基原山麦冬药材鉴别研究

摘要 目的:建立麦冬药材超高效液相-电雾式检测器(UPLC-CAD)特征图谱测定方法,对不同产地麦冬及山麦冬药材进行鉴别研究。方法:采用ACQUITY UPLC BEH Shield RP_(18)(Waters,2.1 mm×150 mm,1.7μm)色谱柱;以乙腈-0.08%甲酸为流动相进行梯度洗脱,流速为0.30 mL/min,柱温为40℃;建立麦冬特征图谱,并对其特征峰进行分析。结果:建立的特征图谱方法符合方法学要求,16批川麦冬及19批浙麦冬样品的特征图谱中各有10个共有峰作为特征峰,通过质谱指认及对照品确认,川麦冬指认了其中5个,浙麦冬指认了其中2个。通过麦冬皂苷元-3-O-α-L-吡喃鼠李糖基-(1→2)-β-D-吡喃葡萄糖苷可区分麦冬和山麦冬;麦冬皂苷D可区分浙麦冬和川麦冬;麦冬龙脑苷可区分湖北麦冬和短葶山麦冬。结论:建立的特征图谱能有效区分川麦冬、浙麦冬及湖北麦冬、短葶山麦冬,该方法具有快速、准确和操作简单等特点,为麦冬药材的快速鉴别提供理论依据。 Objective:To establish a method for the ultra-performance liquid chromatography-charged aerosol detection(UPLC-CAD)of Ophiopogonis Radix,and to identify Ophiopogonis radix from different origins,and its Fakes Liriopes Radix.Methods:ACQUITY UPLC BEH Shield RP_(18)(Waters,2.1mm×150mm,1.7μm)chromatographic Column was used;The mobile phase was acetonitrile-0.08%formic acid,gradient elution;The flow rate was 0.30mL/min.Column temperature was 40℃.Establishing the characteristic spectra of Ophiopogonis Radix to distinguish Ophiopogonis Radix separately form Sichuan Province and Zhejiang Province,with Liriopes Radix.Results:The established characteristic chromatogram method met the method requirements.Ten common peaks were characteristic peaks in the characteristic chromatograms of 16 batchs of Ophiopogonis Radix form Sichuan Province.The characteristic peaks were characterized by UPLC-Q/TOF-MS and the reference standards,and five components were indentified.Ten common peaks were characteristic peaks in the characteristic chromatograms of 19 batchs of Ophiopogonis Radix form Zhejiang Province,Determine the two components.Ophiogenin 3-O-α-L-rhamnopyranosyl-(1→2)-β-D-glucopyranoside can distinguishe Ophiopogonis Radix and liriopes radix.Ophiopogonin D can distinguish Ophiopogonis Radix from Sichuan Province and Zhejiang Province.Borneol 7-O-[β-D-apiofuranosyl-(1→6)]-β-D-glucopyranoside can distinguish the roots from Liriope spicata(Thunb.)Lour.var.prolifera Y.T.Ma and Liriope muscari(Decne.)Baily.Conclusion:The UPLC-CAD method can be effectively distinguished Ophiopogonis Radix from different origins and Liriopes Radix.The method is fast,accurate and simple.It provides a technical basis for the identification of Ophiopogonis Radix.
