中国中南部G8P[8]型A组轮状病毒全基因组遗传特征研究 被引量:1

摘要 G8基因型轮状病毒已在我国出现并迅速增加。本研究拟对采集于中国中南部5岁以下急性胃肠炎患儿的7例G8P[8]型RVA毒株进行全基因组分子特征分析。对轮状病毒特异性扩增结合高通量测序方法对中国中南部14株G8P[8]型RVA毒株进行全基因组测序。利用MEGA11.0、Geneious Prime、Geneious9.0.2和DNASTAR软件对RVA毒株进行全序列及遗传变异和进化分析。7株G8P[8]型RVA的基因分型为G8-P[8]-I2-R2-C2-M2-A2-N2-T2-E1-H2,显示为DS-1样基因型。系统进化树结果显示,本研究中国中南部7株G8P[8]分为两大谱系G8-I(13株)和谱系G8-Ⅱ(1株),VP7的最大分支可信度(MCC)树表明,G8的平均进化率估计为1.4×10^(-3)替换/位点/年。G8P[8]基因型RVA在我国包括两个谱系,从该病毒在中国的出现到该病毒在中国的流行传播将经过1~2年,持续的全国RVA监测至关重要。 Rotaviruses of the G8 genotype have emerged and increased rapidly in China.We undertook genomewide molecular characterization of seven G8P[8]‐type RVA strains collected from children under 5 years of age with acute gastroenteritis in south‐central China.Rotavirus‐specific amplification combined with high‐throughput sequencing was used to sequence the whole genome of 14 G8P[8]‐type RVA strains from south‐central China.Whole‐sequence and genetic variation and evolutionary analyses of the RVA strains were carried out using MEGA11.0,Geneious Prime,Geneious9.0.2 and DNASTAR.Seven G8P[8]‐type RVA strains were genotyped as G8‐P[8]‐I2‐R2‐C2‐M2‐A2‐N2‐T2‐E1‐H2,which was shown to be a DS‐1‐like genotype.The phylogenetic tree showed that the seven strains of G8P[8]in south‐central China were divided into two major lineages:G8‐I(13 strains)and lineage G8‐II(1 strain).The maximum branching confidence(MCC)tree of VP7 indicated that the average evolutionary rate of G8 was estimated to be 1.4×10^(-3) substitutions/locus/year.In summary,the genotype of G8P[8]and RVA comprised two lineages in China.The average evolutionary rate was estimated to be 1.4×10^(-3) substitutions/location/year.The time of emergence to the epidemic spread of the virus in China will take 1–2 years,and continuous national surveillance of RVA is essential.
