干酪乳杆菌ATCC334抑制胶原蛋白诱导关节炎大鼠的炎性反应

Study on Lactobacillus casei ATCC334 Inhibiting Inflammatory Response in Collagen-induced Arthritis Rats

摘要 目的研究干酪乳杆菌ATCC334是否抑制胶原蛋白诱导的关节炎(collagen-induced arthritis,CIA)大鼠的炎性反应以及NF-κB信号通路是否参与了此过程。方法21只Wistar大鼠随机分成3组:A为正常对照组,B为CIA组,C为干酪乳杆菌ATCC334组。组处理因素如下:(1)对照组用生理盐水灌胃,每日500μL,连续28 d不间断;(2)CIA组在造模同时用生理盐水灌胃,每日500μL,连续28 d不间断;(3)ATCC334组造模同时每日500μL浓度为2×10^(8) CFU/mL干酪乳杆菌ATCC334溶解液灌胃,连续28日不间断。应用足爪足趾容积仪测量大鼠后足爪增长容积,第29天处死大鼠,取大鼠血清ELISA检测大鼠血清中TNF-α及IL-6水平,取大鼠腘窝和腹股沟淋巴结制备单细胞悬液分离CD4+T细胞,Western Blot检测CD4+T细胞中TNF-α、IL-6及NF-κB蛋白水平。结果干酪乳杆菌ATCC334降低CIA大鼠关节炎指数及后足爪增长容积,降低了CIA大鼠血清TNF-α、IL-6水平,降低了CIA大鼠CD4+T细胞中NF-κB p65蛋白的表达。结论干酪乳杆菌ATCC334具有抗关节炎和抑制炎性因子的作用,且NF-κB可能参与此过程。 Objective To explore whether Lactobacillus casei ATCC334 can inhibit the inflammatory response of collagen-induced arthritis(CIA)rats and whether the NF-κB signaling pathway is involved in this process.Methods 21 Wistar rats were randomly divided into three groups:A was the normal control group,B was the CIA group,and C was the Lactobacillus casei ATCC334 group.Treatment factors in group A were as follows:(1)In control group,normal saline was given intragastric administration,500μL daily,for 28 consecutive days without interruption;(2)in CIA group,at the same time of modeling,normal saline was administered by gavage,500μL daily,for 28 consecutive days;(3)in ATCC334 group,at the same time of modeling,500μL of Lactobacillus casei ATCC334 solution was injected into the stomach at a concentration of 2×10^(8) CFU/mL every day for 28 consecutive days.The growth volume of the hind foot of the rats was measured using paw and toe volume instrument,and the rats were killed on day 29.Serum levels of TNF-αand IL-6 were detected by ELISA,and CD4+T cells were isolated by single-cell suspension from popliteal and inguinal lymph nodes.The levels of TNF-α,IL-6 and NF-κB proteins in CD4+T cells were detected by Western Blot.Results Lactobacillus casei ATCC334 reduced the arthritis index and hind paw growth volume of CIA rats,reduced the serum levels of-αand IL-6 in CIA rats,and reduced the expression of NF-κB p65 protein in CD4+T cells of CIA rats.Conclusion Lactobacillus casei ATCC334 has the effect of anti-arthritis and inhibits inflammatory factors,and NF-κB may be involved in this process.
