基于高通量测序的输卵管妊娠患者生殖道菌群研究

摘要 目的探讨输卵管妊娠术前后及正常妊娠者生殖道菌群的结构差异,以便了解生殖道菌群对输卵管妊娠及手术治疗的影响。方法选取输卵管妊娠术前后及正常妊娠者共60例为本次研究对象,其中输卵管妊娠术前为BTP组(n=20),输卵管妊娠术后为ATP组(n=20),并选取同期在本院门诊产检的正常宫内早孕(孕6~7周)女性20例作为(EP组)。采用16S rDNA测序分析三组生殖道菌群的多样性和结构差异,并对肠道菌群功能进行预测。结果三组OTU丰度聚类热图提示,三组研究对象的OTU具有明显的差别,但三组菌群的多样性差异无统计学意义(P>0.05);三组研究对象生殖道菌群以乳酸杆菌内酯丰度最高,且BTP组丰度最高;其次是脆乳杆菌,以EP组丰度最高;LDA分析结果提示:三组研究对象生殖道菌群的种类不一致,提示输卵管妊娠及手术对研究对象生殖道菌群的种类有一定的影响;龙胆科、杆菌科、麻风科、细脉藻科、普雷沃特拉科等细菌群类与输卵管妊娠关系密切。结论输卵管妊娠患者输卵管妊娠术前后和正常妊娠者生殖道菌群种类具有明显的差别,表明生殖道菌群对输卵管妊娠能产生重要的影响。 Objective To explore structural difference of reproductive tract microbiota before and after tubal pregnancy surgery and normal pregnant women,to understand effect of reproductive tract microbiota on tubal pregnancy and surgical treatment.Methods The paper chose 60 cases of tubal pregnancy before and after surgery and normal pregnancy as research subjects,analyzed diversity and structural difference of reproductive tract microbiota of three groups with 16S rDNA sequencing,to predict function of gut microbiota.Results Clustering heatmaps of OTU abundance of three groups showed significant OTU difference among three study subjects,there was no statistically significant difference in diversity of three bacterial communities(P>0.05).The highest abundance of lactobacilli lactones was found in reproductive tract microbiota among three groups,and the highest abundance was found in BTP group,followed by lactobacillus fragilis,with the highest abundance in EP group.LDA analysis results showed,types of reproductive tract microbiota among three study groups were inconsistent,indicating tubal pregnancy and surgery havd some certain effect on types of reproductive tract microbiota among study subjects.Bacterial communities of Gentianaceae,Bacteroideaceae,Leprosidae,Leprosidae,and Prevotellaceae were closely related to tubal pregnancy.Conclusion There is significant difference in types of reproductive tract microbiota between patients with tubal pregnancy before and after tubal pregnancy surgery and normal pregnant women,indicating that reproductive tract microbiota can have important effect on tubal pregnancy.
