基于PPARγ/Nrf2信号通路探讨头穴透刺对脑出血大鼠血肿清除的影响

The Effect of Scalp Acupuncture on Hematoma Clearance in Rats with Cerebral Hemorrhage Based on PPARγ/Nrf2 Signaling Pathway

摘要 目的:基于过氧化物酶体增殖物激活受体γ(PPARγ)/核因子E2相关因子2(Nrf2)信号通路探讨头穴透刺对脑出血大鼠血肿清除的影响。方法:将160只雄性SD大鼠分为空白组、假手术组(颅内注射生理盐水)、模型组(脑出血模型)及干预组(脑出血模型+头穴透刺治疗),每组40只。造模后3 d及头穴透刺治疗结束后检测各组大鼠神经功能、脑组织液中血红蛋白(Hb)及一氧化氮(NO)含量、脑组织中Nrf2、CD_(36)蛋白及mRNA表达量;观察各组脑血肿周围微血管分布并计算微血管直径及密度指数。结果:与假手术组比较,造模后3 d干预组与模型组脑组织Hb及NO水平升高,Nrf2、CD_(36)蛋白及mRNA表达均降低,微血管直径及微血管密度指数均减小(P<0.05)。与造模后3 d比较,治疗结束后干预组神经功能评分、微血管直径及微血管密度指数升高,Nrf2、CD_(36)蛋白及mRNA表达降低,且优于模型组(P<0.05);干预组Hb及NO水平降低,且低于模型组(P<0.05)。干预组与假手术组治疗后微血管直径及微血管密度指数比较,差异有统计学意义(P<0.05)。结论:头穴透刺可促进脑出血大鼠脑组织Nrf2、CD_(36)蛋白及mRNA表达,降低脑组织Hb及NO水平,改善血肿周围微血管血流状态,对脑血肿清除有促进作用。 Objective:To investigate the effect of scalp acupuncture on hematoma clearance in rats with cerebral hemorrhage based on the peroxisome proliferator-activated receptorγ(PPARγ)/nuclear factor E2 associated factor 2(Nrf2)signaling pathway.Methods:One hundred and sixty male SD rats were divided into blank group,sham group(intracranial injection of physiological pressure water),model group(intracerebral hemorrhage model)and intervention group(intracerebral hemorrhage model+scalp acupuncture treatment),with 40 rats in each group.After 3 days modeling and after scalp acupuncture,nerve function,hemoglobin(Hb)and nitric oxide(NO)content in brain tissue fluid,Nrf2,CD_(36) protein and mRNA expression in cerebral tissue were measured.The distribution of microvessels around cerebral hematoma in each group was observed and the diameter and density index of microvessels were detected.Results:After 3 days modeling,compared with sham group,Hb and NO levels increased,Nrf2,CD_(36) protein and mRNA expression levels in cerebral tissue of intervention group and model group decreased,and microvessel diameter and microvessel density index were decreased(P<0.05).After 3 days modeling,neurological function score,microvascular diameter and microvascular density index increased,Nrf2,CD_(36) protein and mRNA expression levels in the intervention group decreased after treatment,and those were better in the model group(P<0.05).The levels of Hb and NO in the intervention group were lower than those in the model group(P<0.05).After treatment,there were significant differences in microvessel diameter and microvessel density index between the intervention group and the sham group(P<0.05).Conclusion:The scalp acupuncture could promote the expression of Nrf2 and CD_(36) proteins and mRNA in cerebral tissue of rats with cerebral hemorrhage,decrease the levels of Hb and NO in cerebral tissue,improve the blood flow state of microvessels around hematoma,and promote the removal of cerebral hematoma.
