摘要
目的 探讨丙戊酸钠在蛛网膜下腔出血早期脑损伤大鼠中的实验研究。方法 将120只健康成年雄性SD大鼠根据随机数字表法分为三组,假手术组、模型组及丙戊酸钠组,各40只。颈内动脉穿刺法建立蛛网膜下腔出血模型,丙戊酸钠组大鼠每隔12 h给予丙戊酸钠(300 mg/kg)腹腔注射。原位末端转移酶标记技术检测蛛网膜下腔出血后皮质的细胞和神经元凋亡情况。结果 假手术组大鼠脑底动脉清晰可见,模型组大鼠脑底面有明显的蛛网膜下腔出血。组内比较:与第1天比较,模型组、丙戊酸钠组第3、4天大鼠改良Garcia评分升高,差异有统计学意义(P<0.05);组间比较:前3天,模型组大鼠改良Garcia评分均低于假手术组,差异有统计学意义(P<0.05);第4天,丙戊酸钠组大鼠改良Garcia评分高于模型组,差异有统计学意义(P<0.05)。模型组大鼠脑皮质阳性细胞占比高于假手术组,差异有高度统计学意义(P<0.01);丙戊酸钠组大鼠脑皮质阳性细胞占比低于模型组,差异有统计学意义(P<0.05)。结论 丙戊酸钠减少蛛网膜下腔出血后细胞和神经元的凋亡,改善神经损伤和功能障碍。
Objective To investigate the effect of sodium valproate in rats with brain injury at early stage of subarachnoid hemorrhage.Methods A total of 120 healthy adult male SD rats were divided into sham operation group,model group,and sodium valproate group according to the random number method,with 40 rats in each group.Rats in sodium valproate group were given intraperitoneal injection of sodium valproate(300 mg/kg) every 12 hours.The apoptosis of cortical cells and neurons after subarachnoid hemorrhage was detected by TUNEL staining.Results The basal cerebral artery was clearly visible in the sham operation group,and the subarachnoid hemorrhage was obvious in the model group.Intra-group comparison:compared with the first day,the modified Garcia score of rats in model group and sodium valproate group at 3and 4 days was increased,the differences were statistically significant(P<0.05).Comparison between groups:in the first 3days,the modified Garcia score of rats in the model group was lower than that in the sham operation group,(P<0.05);on day 4,the modified Garcia score of rats in sodium valproate group was higher than that in model group,and the difference was statistically significant(P<0.05).The proportion of positive cells in the cortex of rats in the model group was higher than that in the sham operation group,the difference was highly statistically significant(P <0.01);while the proportion of positive cells in the cortex of rats in the sodium valproate group was lower than that in the model group,the difference was statistically significant(P<0.05).Conclusion Sodium valproate can reduce the apoptosis of cells and neurons,and improve nerve injury and dysfunction after subarachnoid hemorrhage.
