摘要
目的为了研究铁皮石斛根在不同栽培基质下的基因表达差异和调控通路。方法对栽培于碎松树皮、泥炭土等7种基质中的铁皮石斛根部进行转录组测序并比较分析。结果从全部样本中共获得110853条单基因(unigene),其中100194个单基因在Nr、COG等7个数据库中获得注释。与对照相比,栽培于碎松树皮、泥炭土、水苔、碎花生壳、刨花、椰糠和森林腐叶土中的铁皮石斛根分别有55318、55281、39952、58946、25592、52141和56087个差异表达基因。结论实验说明不同栽培基质对铁皮石斛根部的基因表达有较大影响,这些差异表达基因可能是铁皮石斛适应不同栽培基质的重要分子机理,对深入研究铁皮石斛人工栽培技术提供参考和帮助。
Objective To understand the information of differentially expressed genes of different cultivation substrates in Dendrobium officinale Kimura et Migo.Methods The transcriptome of plant under pine bark,peat bog,sphagnum,peanut shell,wood shavings,coir,and forest humus were sequenced by Illumina sequencing technology.Results The results showed that there were 110853 unigenes obtained,among which 100194 unigenes were annotated by seven databases.There were 55318,55281,39952,58946,25592,52141,and 56087 responsive genes in pine bark,peat bog,sphagnum,peanut shell,wood shavings,coir,and forest humus in D.officinale.Conclusion The different cultivation have great influence on gene expression in the root of D.officinale.Differences in gene expression may contribute to the observed mechanism of different cultivation responsive in D.officinale.
