摘要
将2株虫草内生真菌进行发酵培养,进一步研究虫草内生真菌的超氧化物歧化酶(SOD)提取工艺。以酶活力为指标,采用正交实验的方式对分离所得的2株虫草内生真菌(01号和02号)的SOD提取工艺进行研究。实验结果表明:01号菌SOD最佳提取工艺为超声波提取时间45 min,液料比40 mL/g,提取液pH 7.0,所得到的SOD活力为421 U;02号菌最佳提取工艺为超声波提取时间75 min,液料比30 mL/g,提取液pH 8.0,所得到的SOD活力为362 U。通过对比01和02号菌,可知虫草内生真菌中含有丰富的SOD,由于2个产地不同,其虫草内生真菌SOD活力差异较大,提取工艺也有明显差异。
Two strains of Cordyceps endophytic fungi were fermented and cultured to study the extraction process of superoxide dismutase(SOD)from the Cordyceps endophytic fungi.The SOD extraction technology of 2 strains of Cordyceps endophytic fungi(No.01 and No.02)isolated from Cordyceps sinensis was studied by orthogonal experiment with enzyme activity as index.The experimental results showed the best extraction process of SOD of strain No.01 was as follows:ultrasonic extraction time of 45 min,liquid-solid ratio of 40 mL/g,pH of 7.0,and the activity of 421 U.The optimal extraction process of strain No.02 was as follows:ultrasonic extraction time of 75 min,liquid-solid ratio of 30 mL/g,pH of 8.0,and the activity of 362 U.By comparing No.01 and No.02,it can be concluded that Cordyceps endophytes contain abundant SOD.Due to the two different origins,the SOD vitality of Cordyceps endophytic fungi varied greatly,and the extraction process was also substantial different.
