紧密连接蛋白介导的十三味辛夷滴鼻剂对小鼠鼻腔黏膜修复机制探究

摘要 目的 探讨十三味辛夷滴鼻剂对鼻腔黏膜损伤小鼠的修复作用及其可能的干预机制。方法 制备小鼠鼻腔黏膜热损伤模型,随机分为空白对照组、模型对照组、滴通鼻炎水阳性对照组[1μl/(只·次·侧)]、十三味辛夷滴鼻剂高剂量[5μl/(只·次·侧)]、中剂量[2.5μl/(只·次·侧)]、低剂量组[1.25μl/(只·次·侧)],每组各10只,雌雄各半。给药7 d后,分级评价鼻黏膜红肿程度,苏木素-伊红(HE)染色观察鼻黏膜组织病理结构改变,免疫组化分析鼻黏膜上皮ZO-1、Claudin-1、Occludin表达水平。结果 给药7 d后,模型对照组红肿程度评分显著高于空白对照组(P <0.05),滴通鼻炎水阳性对照组、十三味辛夷滴鼻剂高、中、低剂量组均低于模型对照组;HE染色显示,十三味辛夷滴鼻剂各组随剂量增高,黏膜层逐渐变薄、黏膜结构趋于整齐;与模型对照组比较,十三味辛夷滴鼻剂高剂量组ZO-1、Claudin-1、Occludin蛋白表达量显著提高(P <0.05)。结论 十三味辛夷滴鼻剂可能通过上调鼻黏膜上皮紧密连接蛋白ZO-1、Claudin-1、Occludin的表达,修复小鼠鼻腔黏膜损伤。 Objective To explore the repair effect and possible intervention mechanism of Shisanwei Xinyi nasal drops on mice with nasal mucosal injury.Methods A mouse model of nasal mucosal heat injury was prepared and a blank control group,a model control group,and a positive control group of Ditong Biyan Shui(1 μl/[per mouse per time per side]),Shisanwei Xinyi nasal drops high-dose group(5 μl/[per mouse per time per side]),medium-dose group(2.5 μl/[per mouse per time per side]),and low-dose group(1.25 μl/[per mouse per time per side]),with 10mice in each group,half male and half female.After 7 days of administration,the degree of nasal mucosal redness and swelling was graded and evaluated.Hematoxylin-eosin(HE) staining was used to observe the pathological structural changes in nasal mucosal tissue.Expression levels of ZO-1,Claudin-1,and Occludin in the nasal mucosal epithelium were semi-quantitatived by immunohistochemical analysis.Results After 7 days of the administration,the score of redness and swelling in the model control group was significantly higher than that in the blank control group(P < 0.05),while that of the positive control group of Ditong Biyan Shui and the high,medium,and low dose groups of Shisanwei Xinyi nasal drops were all lower than that of the model control group.HE staining showed that as the dosage increased,the mucosal layer gradually became thinner and the mucosal structure tended to be neat in each group of Shisanwei Xinyi nasal drops.Compared with the model control group,the expressions of ZO-1,Claudin-1,and Occludin proteins of the high-dose group of Shisanwei Xinyi nasal drops significantly increased(P < 0.05).Conclusion Shisanwei Xinyi nasal drops repair the nasal mucosal injury of mice by upregulating ZO-1,Claudin-1,and Occludin expression levels in the nasal mucosa epithelium.
