摘要
心血管疾病(cardiovascuar diseases,CVDs)的死亡率位居首位,严重威胁人类健康。沉默信息调节因子5(silent information regulator 5,SIRT5)作为一个新型的去酰基化修饰酶,广泛分布于心肌细胞、血管平滑肌细胞、内皮细胞等心血管细胞中,可通过脱乙酰化、脱琥珀酰化和脱丙二酰化而在多种CVDs中发挥重要作用。该文将从能量代谢、调控炎症反应和氧化应激等方面讨论SIRT5的病理生理学机制,以及SIRT5在心肌梗塞、心肌肥大、心律失常、动脉粥样硬化和心力衰竭等CVDs中所扮演的角色,并且总结了目前SIRT5相关抑制剂和激动剂的研究进展,从而为靶向SIRT5提供防治CVDs的策略。
Cardiovascular diseases(CVDs)are the leading cause of death worldwide and pose a serious threat to human health.Silent information regulator 5(SIRT5),which is widely distributed in cardiac myocytes,vascular smooth muscle cells and endothelial cells,as a novel deacylation-modifying enzyme,plays an important role in CVDs through deacetylation,desuccinylation and demalonylation.This review summarizes the pathophysiological mechanism of SIRT5 from the aspects of energy metabolism,regulation of inflammatory response and oxidative stress,apart from the role of SIRT5 in CVDs such as myocardial infarction,myocardial hypertrophy,arrhythmia,atherosclerosis and heart failure.This review also figures out the current research progress of SIRT5-related inhibitors and agonists,so as to provide strategies for targeting SIRT5 to prevent and treat CVDs.
