LIPC-514C/T多态性与运动干预PDM人群血糖及健康体适能疗效的关联研究

Association between LIPC-514C/T Polymorphism with the Effects of Exercise Intervention on Blood Glucose and Health Physical Fitness in PDM Population

摘要 目的:研究肝脂肪酶基因LIPC-514C/T多态性与糖尿病前期(pre-diabetes mellitus,PDM)人群血糖及健康体适能变化运动干预敏感性的关联,探讨LIPC-514C/T位点作为糖代谢异常人群运动敏感性相关分子标记的可能性。方法:招募32名PDM患者,进行12周中等强度有氧运动结合抗阻训练运动干预。运动干预前、后测试空腹血糖(fasting blood glucose,FPG)、糖化血红蛋白(HbA1c)、体重、身体质量指数(body mass index,BMI)、肌肉重量、内脏脂肪重量、VO_(2max)、握力、纵跳、俯卧撑、坐位体前屈等指标,应用SNP微阵列芯片对唾液LIPC-514C/T位点进行解析,对比运动干预对LIPC-514C/T位点不同基因型PDM患者上述指标的影响。结果:1)LIPC-514C/T位点CC型基因频率为37.5%,CT型为50.0%,TT型为12.5%,基因频率分布符合哈迪-温伯格平衡(P>0.05)。2)运动干预前,LIPC-514 C/T位点不同基因型的PDM人群血糖和健康体适能指标无统计学差异(P>0.05)。3)运动干预后,FPG、HbA1c、体重、BMI、内脏脂肪重量均较干预前显著下降(P<0.05),肌肉重量、VO_(2max)、握力、纵跳、俯卧撑、坐位体前屈均较干预前显著增加(P<0.05)。4)运动干预前、后LIPC-514 CC基因型PDM人群FPG、体重、BMI、肌肉重量、内脏脂肪重量、握力的变化值显著大于CT/TT型(P<0.05),其他指标的变化值在不同基因型间无差异(P>0.05)。结论:12周中等强度有氧结合抗阻训练可改善PDM人群血糖和健康体适能水平,LIPC-514C/T位点CC基因型PDM人群的运动干预敏感性优于CT/TT基因型人群,LIPC-514C/T位点可作为评价糖代谢异常人群运动干预疗效的分子标记。 Objective:To study the association between liver lipase gene LIPC-514C/T polymorphism with the exercise intervention sensitivity to blood glucose and health physical fitness in pre-diabetes mellitus(PDM)population,and to explore the possibility of LIPC-514C/T locus as a molecular marker associated with exercise sensitivity in population with abnormal glucose metabolism.Methods:32 PDM patients were recruited for 12 weeks exercise intervention consisting of moderate intensity aerobic exercise combined with resistance training.Fasting blood glucose(FPG),glycosylated hemoglobin(HbA1c),weight,BMI,muscle weight,visceral fat weight,maximal oxygen uptake(VO_(2max)),grip strength,longitudinal jumping,push-ups,sit forward bend were measured before and after exercise intervention.The LIPC-514C/T locus in saliva was analyzed with SNP microarray chip.The effects of exercise intervention on the above indexes in PDM patients with different LIPC-514C/T locus genotypes were compared.Results:1)The gene frequency of CC genotype in LIPC-514C/T locus was 37.5%,CT genotype was 50.0%,and TT genotype was 12.5%.The gene frequency distribution was consistent with Hardy-Weinberg equilibrium(P>0.05).2)Before exercise intervention,healthy physical fitness indexes,FPG,HbA1c were no statistical difference between different genotypes of LIPC-514C/T locus in PDM patients(P>0.05).3)After the exercise intervention,FPG,HbA1c,weight,BMI and visceral fat weight were significantly decreased compared with those before exercise intervention(P<0.05),while muscle weight,VO_(2max),grip strength,longitudinal jump,push-up,sit forward bend were significant‐ly increased compared with those before exercise intervention(P<0.05).4)The changes of FPG,weight,BMI,muscle weight,visceral fat weight,grip strength of LIPC-514 CC genotype in PDM population before and after exercise intervention were significantly greater than those of CT/TT gen‐otype(P<0.05),there was no difference in the changes of other indexes among different genotypes(P>0.05).Conclusions:12 weeks moderate intensity aerobic combined resistance training can im‐prove the blood glucose level and health physical fitness of PDM population.The exercise interven‐tion sensitivity of CC genotype of LIPC-514C/T locus in PDM population is better than that of CT/TT genotype.The LIPC-514C/T locus can be used as a molecular marker to evaluate the ef‐fects of exercise intervention in people with abnormal glucose metabolism.
